摘 要:目的探讨脑出血(ICH)后出血灶周组织及血清IL-1、IL-10含量的动态变化,分析其与神经元凋亡的关系。方法健康家犬30只,在DSA介入下刺破大脑中动脉制作基底节区或颞叶出血模型,选择出血后6h、12h、24h、48h、72h及96h共6个时间点,每点5只,均于血肿模型制作前及相应时间点处死前检测血清IL-1、IL-10含量,并于各时间点处死后取脑,检测出血灶周组织IL-1、IL-10含量及凋亡神经元的表达。结果ICH灶周组织IL-1、IL-10含量分别于48h及72h达峰值,与相应的血清含量呈正相关(P<0.05),至96h,两者含量均未见下降;灶周凋亡神经元的表达与IL-1含量同步,两者呈正相关(P<0.05)。结论IL-1、IL-10参与了ICH后灶周神经元凋亡的过程,前者诱导或加重了神经元凋亡,后者参与了抑制凋亡的过程,但显示强度不足。Objective To explore the dynamic changes of IL-1 and IL-10 contents in tissue around hemorrhagic focus and serum aider intra-cerebral hemorrhage (ICH), and to analyze the relation between the changes and neuronal apoptosis. Methods Thirty healthy domestic dogs were pricked on middle cerebral artery with DSA intervention, thus dog models of ICH of basal segment region or temporal lobe were prepared. Six time points of 6, 12, 24, 48, 72 and 96 h were chosen, and 5 dogs at each time point were sacrificed. Contents of IL-1 and IL-10 in serum were determined before model preparation and at the 6 time points before sacrification. At each time points aider sacrification, the brain was taken out and the contents of the two substances mentioned above in tissue around hematoma were determined, and the expression of apoptotic neurons was detected too. Results Contents of IL-1 and IL-10 in tissue around hematoma reached a peak at 48 h and 72 h respectively, and there was a positive correlation between the contents of the two substances in tissue around hematoma and those corresponding ones in serum (all P〈0.05). Both contents were not decreased at 96 h. The expression ofapoptotic neurons was synchronous with the level of IL-1 and there was a positive correlation between them (P〈0.05). Conclusion Both IL-1 and IL-10 participate the neuronal apoptosis process around hemtoma, that is, the former induces or aggravates neuronal apoptosis, and the latter suppresses the process of cell apoptosis, but the inhibition is deficient.
