摘 要:目的:研究树突状细胞(DC)与结直肠癌(Lovo)细胞融合疫苗的制备及其生物学特性。方法:①应用PEG融合技术融合树突状细胞和Lovo细胞,制备融合疫苗。②通过描记细胞生长曲线、倒置显微镜和激光共聚焦显微镜、流式细胞仪检测观察细胞生物学特性。③通过检测疫苗的克隆形成率、促进T淋巴细胞增殖能力和疫苗的成瘤活性来观察疫苗的安全性和免疫学效应。结果:融合疫苗呈悬浮生长,具有两种亲代细胞的形态结构特点,CD80、CD83、CD86等在融合细胞表面的表达较单纯DC显著丰富;融合细胞不出现明显的细胞倍增;融合疫苗在软琼脂上培养21 d,未见明显的细胞克隆形成;通过3H-TdR掺入法检测融合细胞刺激T淋巴细胞增殖情况,发现融合疫苗可强烈刺激T淋巴细胞增殖;融合疫苗接种于裸鼠体内观察30 d,未见肿瘤组织生长。结论:融合细胞具有作为抗肿瘤疫苗应具备的低增殖能力和不成瘤的安全性;融合疫苗可明显促进T淋巴细胞的增殖能力说明将其作为抗结直肠癌特异性免疫治疗方法有进一步研究的意义。Objective: To investigate the preparation of the fusional vaccine with dendritic cells and colorectal cancer cells, and its the biological characteristics of the anti-tumor vaccine. Methods:The fusional vaccine was made in the presence of PEG. The biological characteristics of fusion cells , including their morphological specialities, the expression of cell surface phenotypes, growth curves, the ability to irritate cytotoxic T lymphocytes, whether they can proliferate to be a new carcinoma in nude mice etc, were tested to verify their safety when be used as anti-tumor immune reactions. Results: Dendritic cells and Lovo cells could be fusioned by PEG. The fusion cells exhibited typical characteristics by microscope testing ;Their could not proliferate to be new carcinomas in nude mice, but could strongly irritate the proliferation of T lymphocytes. Conclusion : The fusion cells had typical morphologies and are safe as a anti-tumor vaccine in vivo and vitro.
