摘 要:目的探讨盐酸可乐宁对急性视网膜缺血兔玻璃体谷氨酸(glautamate,Glu)浓度的影响。方法在所有动物中,通过升高右眼眼压至120mmHg(1kPa=7.5mmHg)45min建立视网膜缺血模型,左眼为对照眼。药物处理组在缺血前0.5h腹腔内注射盐酸可乐宁0.5mg·kg-1。于缺血后1d和3d收集玻璃体,存放于-80℃冰箱中,高压液相色谱仪测定Glu浓度。结果在对照组中,缺血后第1d、3d时,右眼的玻璃体Glu浓度分别为(47.27±24.98)μmol·L-1、(45.58±7.10)μmol·L-1,与左眼玻璃体谷氨酸浓度相比显著升高(P<0.05)。而在可乐宁处理组尽管在缺血后3d略升高,浓度为(19.11±4.03)μmol·L-1,但并无显著的统计学差异。结论兔急性视网膜缺血可致玻璃体Glu浓度升高;而盐酸可乐宁可以阻止Glu的过量释放。Objective To study and discussthe effect of clonidine on glutamate(Glu) concentration in rabbit vitreous body during acute retinal ischemic insult, Methods In all animals,acute retinal ischemia models were induced in the right eyes by increasing intraocular pressure to 120mmHg ( 1 kPa = 7.5 mmHg)for 45 minutes; the left eyes were sham-operated as the control group. The treatment group were injected intraperitoneally clonidine hydrochloride (0.5 mg·kg^-1)before ischemia. The vitreous was collected separatly on the I st day and the 3rd day after ischemia and stored in a frozen condition ( -80℃). Glu levels were analyzed by high performance liquid chromatography. Results In the control group, the Glu concentration in right eye vitreous was (47.27±24.98)μmol·L^-1 and (45.58± 7. 10)μmol·L^-1 respectively, on the Ist day and the 3rd day after ischemia, significantly higher compared with the left eye(P〈0.05). While the vitreal Glu level wasn't increased significantly in clonidinetreatment group on the 3rd day after ischemia, and the concentration was (19.11±4.03)μmol·L^-1 without statistical difference. Conclusion Acute retinal ischemic damage can result in vetreal Glu concentration increasing; while clonidine hydrochloride can prevent the over release of Glu.
