摘  要:目的应用显微傅里叶变换红外光谱(显微FTIR)技术研究正常大鼠视网膜组织的化学组成。方法在显微FTIR仪下选取正常Wistar大鼠视网膜冰冻切片上同一部位的感光细胞外节、外核层和内丛状层,分别测定反射吸收光谱。通过分析各化学基团振动吸收峰的来源和吸收强度,研究视网膜此三层结构的化学组成及差异。结果外节层CH2基团(2925cm-1、2853cm-1)、CO基团(1738cm-1)和CH基团(3014cm-1)的吸收值均最高。外核层酰胺Ⅰ带(1660cm-1)、PO-2基团(1243cm-1和1089cm-1)吸收值最大。内丛状层CH2、CO和CH基团的吸收介于前两者之间,而酰胺Ⅰ带、PO-2基团吸收最少。结论显微FTIR技术可原位分析视网膜组织,在分子水平为其化学组成提供了大量可靠的信息。Objective To identify the chemical composition of normal rat retinal layers with the technique of Fourier transform infrared (FT-IR) microspectroscopy for exploring a new model system of studies on retinal diseases. Methods Retinal frozen sections of 10 Wistar rat (20 eyes) were analyzed by FT-IR microspectroscopy in reflection mode in the same regions of the retinal outer segments (OS) ,retinal outer nuclear layer (ONL) and retinal inner plexiform layer (IPL). Results There were significant spectral differences among these three layers of retina. OS had the highest absorbance values for CH2 (2 925cm^-1 ,2853cm^-1)(4.357±0.813,1.176±0.226), C=O (1738cm^-1)(3.733±0.814)and =C-H (3014cm^-1) ( 1. 079±0. 264)functional groups due to its composition of stacked membrane disks with high concentration of unsaturated fatty acids. ONL had the highest absorption values for amide I band ( 1 660 cm^-1 ) (42. 679±3. 677) and PO2^- ( 1 243 cm^-1 , 1 089cm^-1) ( 13. 509±3. 794,22.510±3. 861 ) likely due to high concentration of proteins ,DNA and RNA. IPL was found containing concentrations of CH2 , C=O and =C-H functional groups between OS and ONL and the lowest concentrations of amide I band and PO2^- due to the high concentration of synaptic connections. Conclusion FT-IR microspectroscope can be used for in situ characterization of molecular constituents in rat retinal sections. The molecular information obtained from these studies is important in understanding the pathogenesis of retinal diseases.
