摘 要:目的在体外观察K+、Na+对富含端粒重复序列寡核苷酸二级结构的影响。方法在K+、Na+存在下,利用核磁共振光谱仪观察含端粒重复序列的核苷酸d(TTAGGGT)光谱的变化;利用非变性凝胶电泳观察d(TTAGGG)3、d(TTAGGG)4和d(TTAGGG)5迁移速度的变化。结果在含K+或Na+缓冲液中,d(TTAGGGT)在化学移位δ11~12区域内出现代表有鸟嘌呤四分体形成的波峰;K+比Na+具有更强的稳定四链体能力。K+、Na+均可加速d(TTAGGG)4、d(TTAGGG)5在凝胶中迁移速度。结论在体外,K+、Na+可诱导含端粒重复序列的核苷酸形成鸟嘌呤四链体结构。[Objective] To observe the effects of K^+ or Na^+ on the second structure of the ollgonucleotides containing telomeric repeats in vitro. [Methods] In the presence of K^+ or Na^+ , the spectrum changes of the oligonucleotide d (TTAGGGT) containing telomeric repeats were observed using nuclear magnetic resonance (NMR) spectrometer. The mobility of d(TTAGGG)3, d(TTAGGG)4 and d(TTAGGG)5 were surveyed using polyacrylamide gel electrophoresis under non-denaturing condition. [ Results] In the buffers consisting of K^+ or Na^+ , the nucleotide d (TTAGGGT) showed three peaks meaning G-quartet formation within the "chemical shift" field of 11-12 ppm ; and the ability that K^+ stabled G-quadruplex structure was more powerful than that of Na^+. Both K^+ and Na^+ were able to speed mobility of d(TTAGGG)4 and d(TTAGGG)5 in the gel eletrophoresis. [Conclusion] Both K^+ and Na^+ are able to induce the oligonucleotides containing telomeric repeats to form G-quadruplex structure in vitro.
