血凝块DNA提取方法探讨

摘　　要：目的:探讨血凝块DNA提取的方法。方法:利用血凝块融化液直接进行DNA提取,并通过血凝块融化液涂片、血凝块冰冻切片以及琼脂糖电泳鉴定所提DNA的质和量。并与常规研磨法提取DNA进行比较。结果:利用血凝块融化液直接提取DNA浓度可高达(13.9±10.4)mg/L,A(260nm)/A(280nm)为1.85±0.11;高于研磨法的(9.70±4.1)mg/L和1.74±0.08(P<0.05)。血凝块融化液中以及血凝块冰冻切片中均含有较多的有核细胞。结论:利用血凝块的融化液直接进行DNA提取是一种简便、实用的DNA提取方法。Aim： To introduce a novel method for DNA extraction from blood clot. Methods： DNA was extracted directly from the fluid at the bottom of semi-frozen blood clot. The frozen section for the blood clot, smear for blood cells from the fluid and gelose electrophoresis were applied to identify the quantity and quality of the DNA and to explore the possible principle for this method. Results： Rich DNA with high quality could be obtained directly from the blood clot fluid and the obtained DNA concentration was as high as （13.9 ± 10.4） mg/L. Plenty of the nuclear cells were observed on the fluid smear and frozen section. Conclusion： DNA extraction with semi-frozen blood clot is easy and practical.
