摘  要:目的:观察不同材料人工晶状体表面对体外培养的兔晶体上皮细胞黏附、增殖与移行的影响。方法:实验于1999-08/2002-03在辽宁省人民医院中心实验室完成。选取健康家兔10只,双眼正常。按人工晶状体材料的不同分为5组:聚甲基丙烯酸甲酯人工晶状体组14枚,肝素表面处理后聚甲基丙烯酸甲酯人工晶状体组、聚丙烯酸甲酯人工晶状体组、硅凝胶人工晶状体组、水凝胶人工晶状体组各12枚。将体外培养至第3代的兔晶体上皮细胞用质量浓度为1.25g/L的胰蛋白酶消化为单个细胞,用培养液配置细胞密度为1×109L-1的细胞悬液体外接种到各组人工晶状体表面,流式细胞仪检测培养12h时细胞附壁的细胞数,相差显微镜下观察细胞生长情况,并定量分析细胞增殖移行速度。结果:实验纳入聚甲基丙烯酸甲酯人工晶状体14枚,肝素表面处理后聚甲基丙烯酸甲酯人工晶状体、聚丙烯酸甲酯人工晶状体、硅凝胶人工晶状体、水凝胶人工晶状体各12枚,全部进入结果分析。①培养12h各组人工晶状体表面黏附的兔晶体上皮细胞密度比较:聚甲基丙烯酸甲酯人工晶状体组、聚丙烯酸甲酯人工晶状体组均明显高于水凝胶人工晶状体组、肝素表面处理后聚甲基丙烯酸甲酯人工晶状体组、硅凝胶人工晶状体组[(284.75±30.40),(205.00±21.47),(149.17±6.40),(144.33±8.96),(136.17±8.89)个/mm2,P均<0.01]。②兔晶体上皮细胞在不同人工晶状体表面的生长形态观察:各组人工晶状体表面生长的细胞都具有典型的上皮细胞特征,且可见分裂增殖的细胞。③兔晶体上皮细胞在不同人工晶状体表面的增殖移行情况:制造无细胞区后第1天即可看到细胞向心性移行生长,不同材料人工晶状体表面的兔晶体上皮细胞移行速度不同,以硅凝胶人工晶状体组最快,聚甲基丙烯酸甲酯人工晶状体组次之,剩余3组较慢。各组无细胞区内的无�AIM: To observe the adhesion, proliferation and migration of rabbit lens epithelial cells on the surfaces of intraocular lenses of different materials in vitro. METHODS: The experiment was carried out in the central laboratory of Liaoning Provincial Hospital between August 1999 and March 2003. Ten healthy rabbits with normal eyes were used. According to the materials of intraocular lenses,, the experimental rabbits were divided into 5 groups: polymethylmethacrylate (PMMA) group (14 pieces), heparin surface modified polymethylmethacrylate (HSM-PMMA) group (12 pieces), acrylic group (12 pieces), silicone group (12 pieces), hydroxyethylmethacrylate (HEMA) group (12 pieces). 1.0 mL 1×10^9 cells/L aliquots of cultured rabbit lens epithelial cells suspension was seeded onto each lens. The cells adhered on the surfaces of intraocular lenses were digested and counted by flow cytometry at 12 hours. The cell growth was observed under phase contrast microscope, and the cells proliferation and migrated velocity were quantitatively analyzed. RESULTS: All the 14, 12, 12 and 12 pieces of intraocular lenses of PMMA, HSM-PMMA, HEMA, acrylic and silicone were involved in the analysis of results. ① Comparison of the density of rabbit lens epithelial cells at on the surface of intraocular lenses at 12 hours: It was obviously higher in the PMMA group and acrylic group than in the HEMA group, HSM-PMMA group and silicone group [(284.75±30.40), (205.00±21.47), (149.17±6.40), (144.33±8.96), (136.17±8.89)/mm^2, P 〈 0.01]. ② Observation of the growth form of rabbit lens epithelial cells on the surface of intraocular lenses of different materials: The cells growing on the surface of intraoeular lenses in each group all had typical characters of epithelial cells, and splitting and proliferated cells could be observed. ③ Proliferation and migration of rabbit lens epithelial cells on the surface of intraocular lenses of different materials: concentric growth of the
