摘  要:目的:利用小鼠移植瘤模型动态观察CD31、MMP2、MMP9、CathepsinD在恶性黑色索移植瘤血管生成拟态形成中的作用。方法:采用B16黑色素瘤细胞株制作小鼠恶性黑色索移植瘤动物模型,成瘤后根据随机数字表每天随机抽取一只小鼠处死.共12d,获得肿瘤样本114例;HE染色、CD3l、CathepsinD、MMP2、MMP9、HMB45免疫组织化学染色和PAS染色;分别计数血管生成拟态周围及远离血管生成拟态区域肿瘤细胞中阳性细胞百分率;内皮依赖性血管周围及远离内皮依赖性血管区域肿瘤细胞中阳性百分率;连续计数10个高倍视野并取其均值。结果:在恶性黑色素移植瘤形成的第1~8天,肿瘤组织内存在血管生成拟态;第9~12天移植瘤中的血管生成拟态被内皮依赖性血管替代;血管生成拟态周围肿瘤细胞中CD31、MMP2阳性细胞百分率高于远离血管生成拟态区域的阳性细胞百分率;内皮依赖性血管周围肿瘤细胞CD31阳性细胞百分率与远离内皮依赖性血管区域的阳性细胞百分率之间存在明显差别;血管生成拟态周围肿瘤细胞MMP9阳性细胞百分率高于内皮依赖性血管周围的阳性细胞百分率;血管生成拟态密度与血管生成拟态周围肿瘤细胞CD31、CathepsinD、MMP2阳性细胞百分率之间存在相关.与肿瘤细胞中MMP9阳性细胞百分率之问不存在相关,内皮依赖性血管密度只与内皮依赖性血管周围肿瘤细胞中CD31阳性百分率之问存在相关.与CathepsinD、MMP9、MMP2之间不存在相关。结论:恶性黑色素瘤移植瘤中血管生成拟态与内皮依赖性血管共存.且与内皮依赖性血管之问存在一定的时空变化规律。CD31、CathepsinD、MMP2、MMP9、HMB45等参与了黑色素瘤血管生成拟态的形成,其中CD31、MMP2作用尤其重要。Objective: To study the effectiveness of CD31,MMP2,MMPg,CathepsinD on Vasculogenic mimicry in the melanoma transplant tumour. Methods: The mouse melanoma transplant tumor model of mouse was made with B16 melanoma cell suspending liquid. When tumor appears, we choose one mouse each day randomly according to random number chart and kill it. We get tumor specimens of 114 cases totally; which normally will have HE stain CD31, CathepsinD, MMP2, MMP9, HMB45 immunohistochemistry stain and PAS stain. Then respectively count the percentage of positive cells in tumor around the Vasculogenic mimicry and the percentage of positive cells in tumor which is far away from the Vascuolgeni~ mimicry; count the percentage of positive cells in tumor around the endothelial;depended vessel and that of positive cells in tumor which is far away from the endothelial;depended cell. Results: The Vasculogenic mimicry and endothelial-depended vessel co-exlst in the tumor tissue at the early phase of melanoma (d1-d8) ;From day 9 to day 12, this structure is replaced by the endothelial-depended vessel. The number of CD31 ,MMP2 positive cell around vasculogenic miraicry is more than that of the tumor which is far away vasculogenic mimicry. The ratio of CD31 positive cell around endothelial-depended vessel has significant difference with that of tumor far away from it. The number of MMP9 positive cell around vssculogcnic mimicry is more than that of tumor around endothelial-depended vessel. There are corrclation between vasculogenic mimicry density and the number of CD31, MMP2 positive cells around vaseulogenic mimicry, while the density of endothelial vessel has correlation with the number of CD31 positive cell around endothelial depended vessel. But the density of endothelial-depended vessel have no correlation with the number of MMP2 、 MMPg,CathepsinD positive cell around endothelial vessel. Conclusion.
