摘  要:通过现代分子生物学方法,对膀胱癌实体肿瘤的TGFα和P53表达及生物学意义进行了探讨。34例膀胱癌中29例TGFαmRNA表达,表达率为85.3%(29/34);13例癌旁对照组织中,6例无表达,6例低表达,1例高于自身肿瘤表达。肿瘤的表达量是癌旁正常对照的2~5倍,统计学分析,P<0.01。14例膀胱癌4例(28.5%)P53高表达,2例低表达,2例正常和8例癌组织未见表达。膀胱癌TGFα基因的高表达可能是膀胱癌发生发展的早期信号,P53基因的变异表达可能主要参与癌细胞的分化,它可能与膀胱癌恶性表行相关。Transforming growth factor α(TGFα)and P53 mRNA were detected by northern blot analysis in patients with bladde cancer.29 of 34(85.3%) bladder carcinoma expressed TGFαand its level has been 2 ̄5 fold that of normal bladder tissue(p<0.01).The TGFa level was however not relevant to the tumor grade and stage.6 of 14 tumors had P53 mRNA detected,4 with high level and the other 2 with low expression.5 of the 6 P53 positive tumors were of grade 2 or grade 3.7 of 13(54%)normal epithelium tissue adjacent to the tumor has also been TGFαpositive but were of low level except in one.The results denoted that elevated TGFαmRNA might signal an early event in carcinogenesis of bladder cancer while abnormal expression of P53 gene might indicate indirectly the gene mutation and was relevant to the malignancy of bladder tumors.
