转化生长因子β1基因转染对结肠癌SW480细胞增殖的抑制作用

摘　　要：目的：探讨转化生长因子β1（TGF-β1）基因转染对结肠癌细胞SW480体外增殖的抑制作用。方法：以腺病毒为载体将人TGF-β1基因导入体外培养的人结肠癌细胞株SW480，应用免疫组织化学及逆转录聚合酶链反应（RT—PCR）检测SW480细胞中转化生长因子β1（TGF-β1）蛋白及mRNA的表达情况。通过MTT法检测转染后细胞增殖的抑制情况。结果：转染hTGF-β1基因后，与对照组相比，TGF-β1蛋白和mRNA在细胞中的表达均增强。SW480的增殖受到明显抑制（P〈0．05），抑制率为60％～80％。结论：转染hTGF-β1基因对体外培养的结肠癌细胞SW480的增殖有明显抑制作用。Objective: To investigate the effect of infection with adenoviral vector containing human TGF-β1 cDNA on the proliferation of colon carcinoma SW480 cells in vitro.Methods: SW480 cells were infected with recombinant adenovirus containing TGF-β1 in vitro.Protein and mRNA of TGF-β1 were detected by immunohistochemical method and reverse transcriptase-polymerase chain reaction(RT-PCR) technique respectively.The proliferation of SW480 cells was detected by MTT assay.Results: There was increased expression of TGF-β1 in SW480 cell line with transgenosis treatment at protein and mRNA levels.The ability of cell proliferation decreased significantly in SW480 cell line infected with Ad-hTGF-β1,compared with the control group(P<0.05).The inhibition rates arrived at 60%~80%.Conclusion: The proliferation of the colon carcinoma SW480 cell line may be inhibited by transgenosis of TGF-β1 significantly.
