摘 要:目的:观察穹隆-海马伞切断与卵巢切除对大鼠脑内皮质区、海马CA1区、杏仁复合体区、基底前脑Meynert核区胆碱乙酰化转移酶表达的影响,探讨胆碱能损伤和雌激素缺乏对胆碱能神经系统的影响。方法:实验于2003-03/12在青岛大学医学院附属医院脑病防治重点实验室完成。①分组:成年健康雌性Wistar大鼠28只,随机分为双侧穹隆-海马伞切断组、双侧卵巢切除组、双侧穹隆-海马伞切断加双侧卵巢切除组、对照组,每组7只。②模型制备:穹隆海马伞切断动物麻醉固定于脑立体定位仪后,于前囟后2.2~2.5mm,中线外1.0mm处凿开颅骨,切开硬脑膜,自制双刃刀完全切断海马伞外侧缘。双侧卵巢切除动物麻醉后,打开腹腔,在肾脏下方的脂肪内取出卵巢,结扎并切除。对照组除不切断穹隆-海马伞和不切除卵巢外,其余步骤同模型组。③免疫细胞化学染色观察皮质区、海马CA1区、杏仁复合体区、基底前脑Meynert核区等部位胆碱乙酰化转移酶阳性细胞表达的变化。光学显微镜下(10×40倍)计数平均单个视野胆碱乙酰化转移酶阳性细胞的数量。结果:28只大鼠均进入结果分析。①在皮质区,海马CA1区,杏仁核复合体区,Meynert核区胆碱乙酰化转移酶表达阳性细胞数:双侧穹隆-海马伞切断组、双侧卵巢切除组、双侧穹隆-海马伞切断加双侧卵巢切除组均明显低于对照组[皮质区:(2.97±1.45),(3.89±0.65),(4.71±2.81),(32.60±7.33)个/视野;海马CA1区:(6.83±2.41),(4.59±4.50),(3.67±3.53),(50.57±5.85)个/视野;杏仁核复合体区:(14.43±6.75),(13.80±3.63),(15.74±5.72),(35.43±10.49)个/视野;Meynert核区:(5.77±6.62),(7.03±6.77),(0.22±0.37),(48.77±7.10)个/视野,(P<0.01)]。②在Meynert核区,双侧穹隆-海马伞切断加双侧卵巢切除组胆碱乙酰化转移酶表达阳性细胞数明显低于双侧穹隆-海马伞切断组和双侧卵巢切除组(P<0.05)。结论:穹隆-海马伞切断和卵AIM: To observe the influence of fimbria/fornix transection and ovariectomy on the expression of choline acetyltransferase in the cerebral cortex, hippocampus CA1, almond complex and Meynert nucleus of rats. and discuss the effect of cholinergic darnnification and estrogen lack on the cholinergic neurons. METHODS: The experiment was carried out in the Institute of Cerebrovascular Disease, Medical School Hospital of Qingdao University from March to December 2003.①Grouping: Twenty-eight healthy adult female Wistar rats were randomly divided into four groups with 7 rats in each group: bilateral fimbria/fornix transection group, bilateral ovariectomised group, bilateral fimbria/fornix transection +bilateral ovariectomized group and control group. ② Model establishment: The rats were anaesthetized and the bilateral fimbria-fomixes of brain were transected in the stereotaxic apparatus according to Klein's methods. After opening the skull at the coordinates at the bregma anterior-posterior 2.2- 2.5 mm and lateral 1.0 mm according to the Paxinos and so on, the dorsal fornix, the anterior part of hippocampus and the fimbria were cut off with a double blade under visual inspection. About the ovariectomy, the rats were under the condition of anaesthesia, after the abdomen was opened, bilateral ovaries in the fattiness under the kidney were ablated in order to erect the model according to Singh's methods. The animals in the control group were sham-operated by opening the skull only and cut the abdomen only, and the other treatments were the same as those in the model group.③ The changes of expression of the immunoreactive cells of choline acetyltransferase in the cerebral cortex, hippocampus CA1, almond complex and Meynert nucleus were stained by immunohistochemical method. The amounts of the choline acetyltransferase positive neurons in single field were observed under the light microscope (10×40).
