摘 要:目的:观察NogoA及其受体在阿尔茨海默病患者脑内的表达,为研究阿尔茨海默病脑内轴突损伤后不可再生的机制及其治疗提供理论依据。方法:实验于2004-12/2005-09在首都医科大学北京宣武医院老年病研究中心和安徽医科大学第一附属医院完成。由荷兰脑库提供阿尔茨海默病患者3例与非阿尔茨海默病患者3例脑颞叶组织。采用免疫组织化学染色和WesternBlot印迹分析观察Nogo及其受体在两组患者脑海马CA1区和颞叶皮质中的表达。结果:①阿尔茨海默病患者脑颞叶组织中NogoA表达增加而Nogo受体表达减少。②在阿尔茨海默病患者及非阿尔茨海默病患者脑海马组织中,神经元及其突起中都有大量Nogo蛋白的表达;而Nogo受体的表达在阿尔茨海默病患者海马组织中是增加的。结论:①在阿尔茨海默病患者脑颞叶NogoA表达增加,说明阿尔茨海默病患者颞叶有髓纤维损伤严重,可能作为机体的一种保护机制下调了Nogo受体的表达。②在阿尔茨海默病患者脑海马组织中NogoA的表达没有明显改变,但Nogo受体表达却是增加的。这种变化的原因目前还无法解释,可能Nogo受体还有与NogoA无关的其他作用。AIM: To observe the expression of Nogo A and its receptor in the cerebrum of patients with Alzheimer disease (AD), so as to investigate the non-renewable mechamism of internal neuraxis injury in patients with AD as well as provide the theoretical basis for the treatment. METHODS: The experiment was conducted in the Research Center of Geriatrics, Beijing Xuanwu Hospital, Capital University of Medical Sciences and First Affiliated Hospital of Anhui Medical University from December 2004 to September 2005. Cerebral temporal lobe (TL) tissues of 3 patients with AD and 3 patients without AD were supplied by Dutch cerebral storeroom. Expressions of Nogo and its receptor in CA1 region of hip and cortex of temporal lobe in patients were observed with immunestaining and western Blot analysis. RESULTS: ① The expression of Nogo A in cerebral TL tissues of patients with AD increased while the expression of receptor Nogo decreased. ② In the cerebral hip tissues as well as neurons and its ecphymas of patients with and without AD, there were lots of Nogo protein expressions; While the expression of Receptor Nogo was increased in the hip tissues in AD patients. CONCLUSION: ① The expression of-Nogo A in cerebral TL tissues of patients with AD is increased, which indicates that the myelinated fibre of TL in patients with AD are' damaged severely; As a protective mechanism of organism, it may diminish the expression of the receptor of Nogo. ② There are no significant changes in the expression of Nogo A in hip tissues of AD patients, while the expression of receptor Nogo is increased. Reasons of this change is still unknow by now. Perhaps, Nogo has other effects, which doesn't relate to Nogo A.
