摘  要:目的研究核因子κB(NF-κB)及抗凋亡基因Bcl-2在肝外胆管癌组织中的表达及意义。方法应用免疫组织化学SABC法,检测NF-κB在48例肝外胆管癌组织及12例正常肝外胆管组织中的表达变化及抗凋亡基因Bcl-2的相应表达,并分析其与肝外胆管癌临床病理的关系。结果NF-κB在肝外胆管癌组织及正常胆管组织中的阳性表达率分别为75%、16.7%,两者有显著性差异(P<0.01);Bcl-2在肝外胆管癌组织及正常胆管组织中的阳性表达率分别为64.6%、8.3%,两者有显著性差异(P<0.01)。NF-κB的阳性表达在病理分级间有显著性差异(P<0.01)且与Bcl-2的阳性表达显著相关。结论NF-κB是一种与肝外胆管癌有关的癌蛋白,其与Bcl-2高度相关,表明NF-κB可能通过对下游抗凋亡基因表达的调控,参与了肝外胆管癌的抗凋亡过程,影响肝外胆管癌的发生发展。Objective To study NF -κB expression in extrabepatic cholangiocarcinoma tissues and its correlation with the pathological stage of cancer tissues and the expression of Bcl - 2. Methods By means of immunohistochemical SABC method, the expression of NF -κB and Bcl - 2 were compared between the extrahepatic cholangiocarcinoma tissues from 48 cases and normal bile duct tissues from 12 cases. Results The positive expression of NF -κB in the extrahepatic cholangiocarcinoma group and the control group were 75% , 16.7% respectively ( P 〈 0. 01 ). The positive expression of Bcl - 2 were 64.6% ,8.3% respectively ( P 〈0.01 ). The positive of NF -κB was significantly different for different pathological grades(P 〈0.01 ) and the expression of Bcl-2 was closely related to NF -κB ( r = 0. 478 ,P = 0. 001 ). Conclusion NF-κB may be an important oncoprotein, and the abnormal expression of NF -κB is closely associated with the development of extrahepatic cholangiocarcinoma. NF -κB is closely related to Bcl - 2,which indicates that NF -κB is involved in the anti - apoptosis of extrabapotic cholangiocarcinoma cell by mediating anti -apoptosis gene.
