摘 要:目的进一步阐明四氯化碳(CCl4)引起肝纤维化的分子机制。方法用RT-PCR方法检测转化生长因子β1(TGF-β1)mRNA在肝星状细胞(HSC)的表达,用Western blot检测TGF-β1蛋白表达。用MTT法和流式细胞术检测细胞增殖和周期分布。结果CCl4作用于HSC后,细胞的增殖能力增加,细胞周期分布显示G0/G1期细胞减少而G2/M期细胞增多。同时,TGF-β1蛋白及mRNA的表达量明显增加。结论一定浓度的CCl4能够促进HSC增殖,上调TGF-β1蛋白及mRNA的表达,这可能是CCl4导致肝纤维化的分子机制之一。Objective To investigate the molecular mechanism of CCl4 inducing fibrosis in hepatic stellate ceils. Methods The expression of Transforming growth factor-β1 (TGF-β1) at mRNA and protein level were detected by RT-PCR method and Western blot analysis. The proliferation and cell cycle distribution were detected with MTT method and flow cytometry. Results After treatment by 10 mmol/L CCL4 for 24 hours, the mRNA and protein expression of TGF-β1 in CCl4 group sharply increased. In the meantime, the proliferation of CCl4 group ceils was markedly increased. There were less G0/G1 phase ceils and more G2/M phase ceils in CCl4 group than those in control group. Conclusion Suitable CCl4 may increase the expression of TGF-β1 and the proliferation of hepatic stellate ceils, which is one of mechanisms leading to hepatic fibrosis.
