摘  要:目的探讨E26转录因子-1(E26transformation-specific-1,Ets-1)和CD34在视网膜母细胞瘤中的表达及意义。方法利用原位杂交法检测Ets-1mRNA在40例视网膜母细胞瘤中的表达。以免疫组织化学法检测肿瘤血管中CD34的表达,以此计算肿瘤微血管密度的值。结果82.5%(33/40)的视网膜母细胞瘤中Ets-1mRNA呈阳性表达,CD34表达于所有肿瘤组织的血管内皮中,Ets-1主要表达于癌细胞和血管内皮细胞。Ets-1高表达组的微血管密度明显高于Ets-1低表达组(P<0.01)。Ets-1的表达和MVD值与肿瘤的分期存在相关性,随肿瘤分期的增加其表达强度与MVD值均增加。结论Ets-1可促进视网膜母细胞瘤的新生血管形成,可能与肿瘤的侵润和转移有关。Ets-1和CD34的检测可作为视网膜母细胞瘤恶性程度的参考指标。Objective To study the expressions of Ets-1 and CD34 in retinoblastoma(Rb) and their effects on angiogenesis, tumor invasion and metastasis. Methods In sttu hybridization and tmmunohistochemistry (streptomycin avidin-biotinperoxidase complex SP)were used to detect the Ets-1 mRNA and CD34 protein expression in 40 cases of Rb and the microvessel densnty (MVD)was calculated. Results Ets-1 mRNA and CD34 protein were highly expressed in tumor cells with the positive rate of 82.5% (33/40) and 100%, respectively. Ets-1 was over expressed in both Rb cells and endothelial cells.The microvessel density was significantly higher in Ets-1 highly expressed group than in the low expressed group (P 〈 0.01 ). The expression of Ets-1 and MVD showed positive correlation with the tumor's grading.Conclusion Ets-1 could promote angiogenesis in Rb and should be correlated with tumor invasion and metastasis. The detections of Ets-1 and CD34 should be a referred indicator for the angiogenesis and invasion of Rb. [Rec Adv Ophthalmol 2006;26(3):188-190]
