摘 要:应用高效液相色谱-蒸发光散射检测(HPLC/ELSD)系统和微波辅助萃取技术,采用梯度洗脱的方法研究了人参根中人参皂苷的萃取和分离测定;同时研究了人参皂苷含量随人参年龄的变化。结果表明,该方法快速,简单,分离测定准确,可靠。人参皂苷浓度在0.025~0.500g/L的范围内与信号强度值成线性关系;加样回收率为85%-104%;日内精密度≤2.60%;日间精密度≤6.00%。随着人参年龄的增长,人参根中的人参皂苷含量也增加。A method for the determination of six kinds of ginsenosides in the root of radix ginseng, which were successfully separated using gradient elution by high performance liquid chromatography (HPLC) equipped with evaporative light scattering detector ( ELSD), was developed. The extraction of six ginsenosides from radix ginseng root was performed by microwave-assisted extraction (MAE). The MAE procedure provided the enhanced extraction efficiency and the reduced processing time. The column to be used in this study was a cosmosil-C18 reversed-phase, 4.6 mm × 250 mm, 5 μm and the mobile phase was acetonitrile and water. The calibration curves of the six ginsenosides were all linear over the range of 0. 025 - 0.500 mg/L with good correlation coefficients (r 〉 0.99). The recoveries for the ginsenosides were in the range of 85%- 104%. The results proved that the older the radix ginseng, the higher the contents of ginsenosides in the root of radix ginseng.
