摘 要:目的:研究白细胞介素-2(IL-2)与单纯疱疹病毒1型(HSV-1)糖蛋白D(gD)基因共表达的核酸疫苗对小鼠免疫效果的调节作用。方法:构建表达HSV-1gD基因的核酸疫苗IRES-gD及共表达HSV-1gD与IL-2基因的重组质粒IRES-gD-IL-2,给BALB/c小鼠股四头肌注射,检测各组特异性抗体、中和抗体水平以及血清IgG2a/IgG1比值;并测定迟发型超敏反应(DTH)的强度;通过病毒攻击实验观察各组小鼠的生存情况。结果:IRES-gD-IL-2组产生的抗体及中和抗体滴度明显高于IRES-gD组。IRES-gD-IL-2免疫小鼠的IgG2a和IgG1含量及DTH反应均较IRES-gD组明显增高,但两组小鼠的生存率无显著性差异。结论:IL-2基因与HSV-1gD基因共表达可全面增强小鼠对核酸疫苗的免疫应答,尤其可增强细胞免疫效果。Aim: To investigate the regulating effect of interleukin-2 (IL-2) gene on the immune response induced by herpes simplex virus type 1 (HSV-1) glycoprotein D (gD) gene in mice. Methods: Eukaryotic expression plasmid IRES-gD which encoded HSV-1 gD gene and co-expressive nucleic vaccine plasmid of IRES-gD-IL-2 which contained both HSV-1 gD and IL-2 gene were constructed as nucleic vaccines, and then they were transfected to dendritic cell(DC) by using lipofectamine, respectively, and were injected to BALB / c mice via the i.m. routes. The induced antibody titers were determined by ELISA. The DTH response in each mouse was expressed as the difference in thickness between the left and right pinnae, the protective immune responses against HSV-1 challenge were evaluated. Results: Mice vaccined with IRES-gD-IL-2 had much higher IgG antibody titer and neutralizing antibody titer compared with the mice received IRES-gD group. Both IgG2a and IgG1 were increased in the IRES-gD-IL-2 group than those in the IRES-gD group. Moreover, the DTH response was also further increased in the IRES-gD-IL-2 group to a higher level than that in the group with received IRES-gD. Both of groups provided significant protection against lethal HSV-1 challenge, but there was no difference between the two groups. Conclusion: Injection of mice with plasmid DNA coexpression of HSV-1 gD gene and IL-2 gene could enhance both cellular and humoral immune responses efficiently, and achieve better protection against HSV-1 challenge.
