摘 要:采用增殖细胞核抗原(PCNA)免疫组化标记与图像分析技术,对30例人体肝细胞癌与癌旁近癌肝细胞的增殖活性进行了定量研究。结果显示:(1)癌与癌旁近癌肝细胞PCNA阳性平均标记率分别为45.22%±17.90%和12.74%±4.89%(P<0.01),随肝癌分级升高,PCNA标记享有升高的趋势。(2)40%以上肝癌细胞的PCNA相对含量与癌旁肝细胞PCNA平均含量相似,结合病理形态分析,认为癌细胞PCNA含量增高与低分化癌细胞核增大关系密切。(3)用图像分析技术显示的PCNA标记数密度与常规计数法所得PCNA标记率有较好一致性,且前者更为客观。The proliferative:activity of hepatocellular cancerous cells(HCCs)and cancer-adjacent hepa tocytes were quantitatively studied with immunohistochemical assay of the proliferating cell nuclear antigen(PCNA)and computerized image analysis.The findings were as follows:(1)The average rate of PCNA labelling was 45.22%±17.90%and 12.74%±4.89%in HHCs and cancer-adjacent hepatocytes respectively(P<0.01)and the rate of PCNA labelling tended to increase with the increase of the pathological grading of hepatocellular carcinoma,(2)The relative contnet of PCNA in 40%of HCCs was similar to the average content of PCNA of cancer-adjacent hepatocytes.In combination to the histopathological findings,it is concluded that the increase of PCNA content in HCCs was closely related to the increase of their nuclear size,(3)A coincidence was obtained between the numeral density of PCNA-positive cells with image analysis and the labelling rate with routine counting and the former was considered as a more objective index.
