摘 要:目的探讨8-氯腺苷(8-chloroadenonosine)是否对体外培养的晶状体上皮细胞增生有抑制作用。方法取第2、3代传代培养的晶状体上皮细胞做药物抑制试验。加入不同浓度的8-氯腺苷(2,4,8,16μmol/L)作用24、48、72和96小时后,用MTT比色测定法和流氏细胞仪检测法确定药物对兔晶状体上皮细胞增生的抑制作用。结果 8-氯腺苷在4-16μmol/L的浓度下能抑制晶状体上皮细胞的增生。并且其抑制程度呈时间、剂量依赖性。结论该结果可为临床筛选防治后发性白内障的药物提供依据。Objective In order to observe the 8-chloroadenonosine has inhibitor effect on human lens epithelial cell growth. Methods The second or third passage of human lens epithelial cells growth we re used to be treated with different concentration of 8-chloroadenonosine( 2,4,8,16μmol/L. The effect of 8-chloroadenonosine on human lens epithelial cells growth were examined by MTr assay method and Flow cytometer(FCM) after treated for 24h,48h,72h and 96h. Resuits 8-chloroadenonosine can inhibit the growth of human lens epithelial cell at the concentration from 4μmol/L to 16μmol/L. And the effect depend upon the times and concentration. Conclusion The 8-chloroadenonosinc is perhaps helpful in treating posterior capsular opacification after cataract surgery.
