摘  要:目的探讨强迫游泳不同应激阶段和应激强度对大鼠行为和内分泌的影响,并分析这些改变的精神病理学意义。方法建立强迫游泳模型,将大鼠随机分为对照组、单次应激组、2周应激组和4周应激组。采用旷场试验评定应激后大鼠的行为反应,酶联免疫吸附实验检测血清皮质酮水平。结果随着应激次数增加,大鼠的主动性运动逐渐减少,而被动性运动逐步增加。单次应激组的直立和修饰[(7.50±2.04)次,(8.33±1.88)次]明显高于对照组[(1.84±0.35)次,(5.05±0.88)次]、2周应激组[(2.54±0.66)次,(4.21±0.37)次]和4周应激组[(2.83±0.75)次,(3.61±0.46)次]。2周应激组和4周应激组血清皮质酮水平[(1001.35±202.91)ng.ml-1,(728.67±250.87)ng.ml-1]明显高于对照组[(626.52±349.89)ng.ml-1]和单次应激组[(359.09±164.32)ng.ml-1]。血清皮质酮水平与应激强度呈正相关(r=0.488,P<0.05)。应激适应组修饰、直立的次数[(6.11±5.04)次,(5.67±4.92)次]明显高于非适应组[(3.64±1.96)次,(1.64±1.06)次]而其皮质酮水平[(580.43±383.52)ng.ml-1]明显低于非适应组[(812.32±157.80)ng.ml-1]。结论(1)单次应激组大鼠表现焦虑样行为增多,慢性应激可导致大鼠行为的明显抑制;(2)在强迫游泳实验中存在主动性与被动性行为个体差异,具有不同的精神病理学意义。Objective This study investigated the behavior and endocrine changes of forced swimming (FS) stressed rats to evaluate psychopathological implications of animal models. Methods Using FS test as stressor, rats were divided into control, single,2 weeks stressed and 4weeks stressed groups randomly. Before and after stress mission, behaviors were measured by open field test. SerUm eortieosterone level was analyzed by enzyme-linked immunosorbent assay. Results As stress times is increasing, stressed rats showed decreasing active locomotion gradually but increasing passive locomotion. Meanwhile, there were decreasing trends of staying time, crossing"and rearing .Single stressed rats have more rearing and defecating(7.50 ± 2.04,8.33 + 1.88 ) compared with other groups. The serum corticosterone level of 2 weeks and 4 weeks stressed rats [ ( 1001.35 ± 202.91 ) ng· ml^-1, (728.67 + 250.87 )ng· ml^-1 ) ] is reliably higher than other groups. It was positively correlated to stress times ( r = 0. 488, P 〈 0.05 ) . There were more defecating and rearing and low serum eorticosterone level in stress adaptation group(6.11 ± 5.04,5.67± 4.92,580.43 ± 383.52ng· ml -1 ) compared to un-adaptation group( 3.64 ± 1.96,1.64 ± 1.06,812.32 ± 157.80ng· ml - 1 ). Conclusions ( 1 ) Single FS stressed rats mainly show increase of anxiety-like behavior, while chronic stressed rats may induce marked restrain ; ( 2 ) The individual difference of active and passive behavior in FS test has different phyehopathologieal implication.
