摘 要:目的 探讨PTEN-PI3K信号转导途径中PTEN与PI3K的蛋白表达与非小细胞肺癌(NSCLC)生物学行为的关系。方法 采用第二代免疫组化Elivision法和Western blot法检测59例NSCLC组织中PTEN与PI3K的蛋白表达。结果 通过Western blot和免疫组化检测PTEN、PI3K的蛋白表达结果一致,在NSCLC组织中阳性表达率分别为55.93%(33/59)、77.97%(46/59)。NSCLC组中PTEN、PI3K蛋白的阳性表达率分别与正常肺组织和癌旁增生肺组织相比,差异均有显著性(P〈0.05)。PTEN蛋白阳性表达率与肺癌的组织学分型、分化程度无关(P〉0.05),而与淋巴结转移有关(P=0.009)。PI3K蛋白的阳性表达率与NSCLC的组织学类型无关(P〉0.05),但与细胞分化程度以及与淋巴结转移均有关(P〈0.05)。PTEN与PI3K之间具有显著负相关性(P=0.003)。结论 PI3K蛋白的过表达和PTEN蛋白的低表达与肺癌的发生以及肺癌的浸润转移密切相关。PTEN蛋白的低表达可能刺激了PI3K蛋白的强表达。Objective To investigate the relationship between PTEN and PI3K protein expressions of PTEN-PI3K signal transduction pathway and biological behavior of non-small cell lung cancer (NSCLC). Methods PTEN and PDK protein levels in 59 patients with NSCLC were detected by second generation immunohistochemical Elivision methods and Western blot. Results The coincident results of PTEN and PI3K protein expressions were observed by Western blot and immunohistochemistry, PTEN and PI3K proteins positive rates were 55.93% (33/59) and 77.97% (46/59) respectively in 59 NSCLC. Significant differences were both present between PTEN and PI3K proteins positive rates of NSCLC and those of normal lung tissue, para-cancerous hyperplasia lung tissue respectively (P〈0.05). The positive expression of PTEN protein was not associated with the pathological types and cellular differentiation degree (P〉0.05), but highly associated with lymph node metastasis (P=0.009). The positive expression of PI3K protein was not associated with the pathological types (P〉0.05), but highly associated with cellular differentiation degree and lymph node metastasis (P〈0.05).In 59 NSCLC, a negative correlation was present between PTEN and PI3K protein expression (P=0.003). Conclusion Overexpression of PI3K protein and low-expression of PTEN protein are high related with the development, invasion and metastasis of lung carcinoma. Low-expression of PTEN protein may stimulate overexpression of PI3K protein.
