摘  要:目的研究脂膜微囊(LCM)靶向性聚集于大鼠颅内C6胶质瘤动物模型的机制。方法建立大鼠C6胶质瘤动物模型,成瘤大鼠分为两组:单次注射LCM组,多次连续注射LCM组,处死后获取大鼠脑组织标本,进行常规HE染色、油红O染色,荧光显微镜观察LCM在肿瘤位点分布的特点,取C6胶质瘤细胞研究LCM在细胞内的分布特点。结果油红O染色显示肿瘤细胞内有LCM的聚集,且多次注射LCM组其LCM浓度明显较单次注射组高。荧光显微镜观察发现,LCM主要聚集于肿瘤区,且主要分布于肿瘤细胞的胞浆中。对体外培养的C6胶质瘤细胞研究发现,LCM先结合于细胞表面,随后在细胞内再分布,其在细胞内的分布可能与细胞内的酸性成分有关。结论LCM可靶向性聚集于肿瘤位点,其在肿瘤位点浓度与注射次数成正相关。其靶向性机制与LCM的脂质构成有关,还与脑肿瘤引起的局部血脑屏障破坏有关。LCM在肿瘤细胞内的代谢与细胞内的酸性成分有关。Objective To investigate the mechanisms of targeted aggregation of lipid-coated microbubbles (LCM) towards intracranial C6 glioma in rat models. Methods C6 glioma models of rats were established and separated into 2 groups: single tail vein injection of LCM, several consecutive injections of LCM. The models were then sacrificed and the brains were harvested for HE stain and oil red O stain. Immunofluorescent microscope was used to observe the LCM distribution characteristics in tumor sites and in C6 glioma cells, Results Oil red O stain showed that LCM aggregated in tumor cells and the LCM density of multiple injection group was much higher than that of single injection. Fluorescent microscope showed that LCM aggregated mainly in tumor region, and specifically in the cytoplasma of tumor cells. The mechanisms by which LCM entered the tumor cells were investigated in vitro, that was, LCM first connected with the cellular surface, and then redistributed in cells, which was associated with the acidic component of cytoplasma, Conclusion The density of LCM in tumor sites is associated positively with the number of injection. The mechanisms of targeted aggregation are related to the lipid component of LCM, also to the local breakage of blood brain barrier induced by brain tumor. The metabolism of LCM in tumor cells depends on acidic component of cytoplasma.
