摘  要:目的探讨磁流体热处理对小鼠H22移植瘤生长及血管内皮生长因子(VEGF)表达和凋亡状况的影响。方法将接种了H22肝癌细胞的小鼠随机分为4组。对第4组移植瘤内注射葡聚糖磁流体,在55kHz,20kA/m交变磁场中作用10min。第1、第2、第3组作为对照,分别对瘤内注射盐水不加磁场,注射盐水加磁场和注射磁流体不加磁场。两周后处死动物,称瘤重,计算抑瘤率。另两组动物分别瘤内注射磁流体和等体积盐水,作为处理组和对照组,在经磁场处理后1h、5h、24h、48h和336h处死,检测瘤组织的VEGF表达和凋亡状况。结果第4组移植瘤内的温度可达到46℃~48℃。与第1、第2、第3组相比,第4组抑瘤率分别为:59.74%、52.51%和40.35%,其中与第1组差异有显著性(P<0.05)。磁热处理后不同时间,处理组肿瘤组织的VEGF表达和凋亡状况与对照组的比较差异均无显著性。结论单次磁流体热处理可以显著抑制小鼠H22肝癌移植瘤的生长。抑制VEGF表达和诱导凋亡不是抑制移植瘤生长的主要原因。Objective To study the effects of hyperthermia with dextran magnetic fluid on growth, vascular endothelial growth factor (VEGF) expression and apoptosis of grafted H22 tumor in mice. Methods Mice inoculated H22 tumor cells were divided into four groups randomly. The fourth group was injected into tumor with dextran magnetic fluid and treated with 55kHz, 20 kA/m alternating magnetic field for 10 min subsequently. The first, the second and the third group regarded as control groups, were injected with physiological saline, injected with physiological saline and suffered magnetic field, and injected with dextran magnetic fluid respectively. Mice were sacrificed in two weeks. The tumors were weighed to calculate the ratio of inhibition. Other two groups were inoculated as above, the treatment group injected with magnetic fluid and the control group injected with physiological saline. After treated with magnetic field lh, 5h, 24h, 48h and 336h, mice were sacrificed respectively. The VEGF expression and apoptosis of tumors were detected. Results The temperature in the center of tumor in the fourth group ranged from 46℃ to 48℃. Compared with the first three groups, the ratios of inhibition of the fourth group were 59. 74%, 52. 51% and 40. 35% in turn, The difference between the first and the fourth group has statistical significant (P〈0. 05). In different time after treatment, the VEGF expression and apoptosis in tumors of the treatment group were not obviously different from that of control group. Conclusion Single hyperthermia treatment with dextran magnetic fluid inhibited growth of H22 tumor in mice. The main reasons of hyperthermia that inhibit tumor growth were not because of inhibition in VEGF expression and inducing apoptosis.
