摘 要:目的:观察大鼠重度心肌挫伤后不同时间段挫伤区周围心肌细胞中bcl-2的表达情况,从分子水平上进一步探讨心肌挫伤的损伤机制。方法:成年Wistar大鼠36只,随机分为对照组和实验组。建立大鼠重度心肌挫伤模型,运用免疫组织化学技术(SABC法),结合计算机彩色图像分析技术观察大鼠实验性重度心肌挫伤后bcl -2、的染色变化。结果:免疫组化染色观察,bcl-2在正常对照组以及死后损伤组为阴性表达,重度心肌挫伤后, 在挫伤区周围组织bcl-2在6h即出现轻微表达,并分别在24h和2d达到高峰,此后逐渐下降,至6d仍有表达。结论:bcl-2参与了重度心肌挫伤后细胞的损伤;心肌挫伤后的时序性变化规律使bcl-2有可能成为重度心肌挫伤时间推断的客观指标以及生前伤和死后伤的鉴别指标。Object : To observe the expression of bcl-2 staining in myocardial cell round the wound area heart of rat after experimental severe myocardial contuslonand to clarify the molecular mechanism of severe myocardial contusion. Method: A experimental severe myocardial contusion model was established, and using SABC immunohistochemical and computer image analysis, expressions of bcl-2 protein after severe myocardial contusion in different time were evaluated. Results: the control-experiment is negotive, the expressions of bcl-2 and could be observed at 6h after severe myocardial contusion, and then increased with the prolonging of time , then got to a peak at 12h, 12h and 2d after injury respectively, by 6d the positive staining cells 8till could be detected. Conclusion: bcl-2 may play a role in severe myocardial contusion; as an objective indication, the time sequence regularity of bcl-2 and after severe myocardial contusion may be applied in severe myocardial contusion time estimation.
