双黄补对牙周病中牙周膜细胞的增殖分化的调控

摘　　要：目的:探讨中药双黄补对体外培养的人牙周膜细胞(PDLC)增殖和分化的影响。方法:通过体外培养人PDLC,应用双黄补提取液作为辅助因子分成8组:A·黄连;B·黄芩;C·骨碎补;D·黄连加黄岑;E·黄连加骨碎补;F·黄岑加骨碎补;G·黄连、黄芩、骨碎补;H·对照组。采用MTT比色法测定细胞增殖情况,羟脯氨酸法测定胶原蛋白比值。结果:各组药物提取液均有促进PDLC增殖和提高胶原蛋白比值的作用,与对照组比较,差异有显著性,其中以双黄补组的效果最明显。不同孵育时间培养中以第5天的作用最强。结论:双黄补水提液可明显促进人PDLC增殖,明显提高胶原蛋白在总蛋白中的比值,可作为PDLC增殖的理想的辅助因子。Objective：To observe the effects of shuanghuangbu（goldthread, skullcap, rhizome drynafiae） on the proliferation and differentiation of human periodontal ligament celIs（PDLC）. Methods： For the proliferation assay, PDL cells were cultured in different groups with different ingredients, and cell numbers were counted by using MTT method and the ratio of collagen content in total protein was evaluated with hydroxyproline. Results：All groups of different ingredients stimulated the proliferation and differentiation of PDL cells and increased the ratio of collagen content in total protein except goldthread group （A group）. The effect of shuanghuangbu group（G） was the most significant. All results were compared with those in the control group（H） （ P 〈 0.05）. Conclusions： These findings suggest that, as a traditional Chinese herb, shuanghuangbu can significantly stimulate the proliferatin and differentiation of PDL cells, and may play an important role in the regeneration of PDL.
