牛磺酸对大鼠肝星状细胞的影响

摘　　要：目的探讨天然牛磺酸对大鼠肝星状细胞的影响。方法采用HSC-T6细胞株为模式细胞,传代培养,MTT法观察天然牛磺酸对HSC-T6细胞增殖的影响,胶原晶格收缩法观察其对HSC-T6细胞收缩的影响,ELISA法观察其对HSC-T6细胞分泌I型胶原(CoI)、纤维连接蛋白(FN)、转化生长因子-β1(TGF-β1)的影响。结果天然牛磺酸中、高剂量组对HSC-T6细胞抑制率分别为29·72%,33·04%,与对照组相比差异具有统计学意义(P<0·01);各剂量组较对照组均能显著抑制HSC-T6收缩(P<0·05或P<0·01),抑制作用与剂量呈依赖关系;中、高剂量组较对照组相比能显著抑制HSC-T6分泌CoI、FN、TGF-β1(P<0·05或P<0·01)。结论天然牛磺酸可抑制HSC-T6增殖、收缩、分泌CoI、FN、TGF-β1。Objective To investigate the effect of natural taurine on hepatic stellate cells.Methods HSC-T6 was chosen as the study model, cells of this model were subculture. MTT assay was used to evaluate the proliferation of HSC-T6. The HSC-T6 cells contraction were studied by the contraction of collagen lattice. ELISA method was used to evaluated the secretion of collsgen I（Col I）,fibnmectin（FN） and transforming growth factor-β1（TGF-β1）.Results HSC-T6 cells inhibiting rate of natural taurine at mediate-dose group and high-dose group were 29.72% and 33.04%, which were significantly higher than that of control group（ P〈0.01）. All dose groups could obviously inhibit the contraction of HSC-T6 cells compared with control group（ P 〈0.05 or P 〈 0.01）as direct relation of concentrstions.mediate-dose group and high-dose group of natural taurine could inhibit the secretion of CoI,FN,TGF-β1 compared with control group（ P 〈 0.05 or P 〈 0.01）. Conclusions Atural taufine can inhibit proliferation,contraction and excreting CoI,FN,TGF-β1 of HSC-T6.
