Ag85B/IL-2融合蛋白在体外对PBMC增殖及Th1型细胞因子分泌的影响

摘　　要：【目的】研究Ag85B/IL-2融合蛋白对人外周血单个核细胞(PBMC)增殖及Th1型细胞因子分泌的影响。【方法】MTT法测定不同浓度的Ag85B/IL-2融合蛋白在不同时相对人PBMC的增殖活性,ELISA法检测Ag85B/IL-2融合蛋白作用人PBMC不同时间后IL-12和IFN-γ的分泌水平,并与Ag85B和BCG作对比。【结果】以0.8μg/ml Ag85B/IL-2融合蛋白刺激组的人PBMC增殖活性为最强,此组在各时段均显著高于空白对照组(P<0.01,P<0.05);各浓度Ag85B/IL-2融合蛋白刺激48 h后的IL-12水平均显著高于空白对照组(P<0.01,P<0.05),以0.8μg/ml的浓度刺激PBMC 72 h时,IL-12的分泌达最高;0.8μg/ml和1.6μg/ml刺激组的IFN-γ浓度在各个时相点均显著高于空白对照组(P<0.01,P<0.05)。Ag85B/IL-2融合蛋白刺激PBMC分泌IL-12和IFN-γ的作用,均显著强于Ag85B和BCG(P<0.01)。【结论】Ag85B/IL-2融合蛋白具有促人PBMC增殖和Th1型细胞因子分泌的作用。[Objective]To research the in vitro proliferation and Thl type cytokine secretion effect of human peripheral blood mononuclear cells （PBMC）activated by Ag85B/IL-2 fusion protein. [Methods]After activated by different concentrations of Ag85B/IL-2 fusion protein at different hours, the proliferation activity of PBMC was analyzed by assay of MTT, and the Thl type cytokine IL-12 and IFN-γ secreted by PBMC were detected with enzyme-linked immunoabsordent assay（ELISA） and compared with those of stimulated by Ag85B and BCG. [Results]The PBMC proliferation of 0.8μg/mL fusion protein stimulating group was highest and significantly higher than that of blank control group at different stimulating time（ P 〈0.01, P 〈0. 05）. The IL-12 level was significantly higher than that of blank control groups when stimulated for at least 48 h at different stimulating concentrations （ P 〈0.01, P 〈0.05）, and the IL-12 secreting peak was at 0.8 μg/mL concentration for 72 h. The IFN-γ levels of 0.8 μg/mL and 1.6 μg/mL stimulating groups were significantly higher than those of control groups at different time（ P 〈0.01, P 〈0.05） ,and the levels of IL-12 and IFN-γ of Ag85B/IL-2 fusion protein stimulating group were significantly higher than those of Ag85B and BCG stimulating groups（ P 〈0.01）. [Conclusion]Ag85B/IL-2 fusion protein may promote the proliferation of human PBMC and Thl type cytokine IL-12 and IFN-γ secretion.
