摘 要:目的探讨外源性 p14ARF 基因对胰腺癌 PC-3细胞血管生成的影响。方法将转染了外源性 p14ARF 基因的人胰腺癌 PC-3细胞株接种裸鼠形成实体瘤,用 RT-PCR 和 Western blot 法观察 VEGFmRNA 及 VEGF 蛋白的表达,并对实体肿瘤的微血管密度进行检测。结果转染p14ARF 基因后的 PC-3细胞实体瘤中 VEGFmRNA 及 VEGF 蛋白的表达下调,血管密度降低。结论p14ARF 基因能下调内源性 VEGF 的表达,有着抗血管生成的治疗意义。Objective To study the effects of pl4ARF gene on angiogenesis of pancreatic carcinoma cell line PC-3 in vivo. Methods The model of xenografts was established by implanting PG3, PC-3/vec and PC-3/pl4 cells under the skin in nude mice. RT-PCR and Western blot were used to observe the changes in angiogenesis-associated factor, VEGF, in the 3 kinds of cells. Meanwhile, the microvessel density (MVD) was determined with immunohistochemistry using CD34 antibody. Results The expression of VEGF was reduced in PC-3/pl4ARF xenograft, which was in consistent with the decrease in MVD. Conclusions pl4ARF gene can exert anti-angiogenesis effects on PC3 cell line by down-regulation of VEGF expression.
