摘  要:目的探讨大脑中动脉闭塞(MCAO)后早期胰岛素样生长因子-1(IGF-1)mRNA和蛋白的表达。方法应用12800点DNA芯片、原位杂交和免疫组化方法检测SD大鼠MCAO后IGF-1mRNA和蛋白的表达水平。结果基因芯片结果显示MCAO后缺血侧纹状体内有4480个差异表达基因,其中个别生长因子类基因呈高水平表达,但是多数基因呈低水平表达,IGF-1mRNA在脑缺血后呈相对低水平表达。原位杂交和免疫组化法进一步证实,与假手术组相比,MCAO后缺血侧纹状体内IGF-1mRNA和蛋白阳性细胞数明显减少,再灌注9h和24h时表达均降低(P<0·001)。结论MCAO后早期缺血侧纹状体内IGF-1mRNA和蛋白呈低水平表达,提示IGF-1在脑缺血损伤中可能具有一定的作用。Objective: To explore the expression of insulin-like growth factor-1 (IGF-1) mRNA and protein following middle cerebral artery occlusion (MCAO). Methods: The 12 800 dots DNA chips analysis, and in situ hybridization and immnuohistochemistry were applied to detect the expression levels of IGF-1 mRNA and protein in SD rats. Results: The result of gene chip showed that 4 480 differential expressed genes in the ischemic striatum following MCAO, among them individual growth factor family genes were up-expressed, whereas most of them were down-expressed. IGE-1 mRNA was down-expressed following cerebral ischemia. In situ hybridization and immunohistochemistry further confirmed that in comparison with the sham-operation group, the numbers of IGF-1 mRNA and protein immuno-positive neurons significantly decreased in the ischemic striatum after MCAO, and the expression decreased after 9 and 24 hours reperfusion (P 〈 0.001 ). Conclusion: The expression of IGF-1 mRNA and protein in early ischemic striatum following MCAO decreases and this suggests that IGF-1 may play a role in cerebral ischemic injury.
