摘 要:目的:探讨热激蛋白(heat shock protein,HSP)90、70、27在高转移性乳腺癌细胞株MDA-MB-435s和MDA-MB-231中的表达情况及其意义。方法:MTT法检测HSP 90抑制剂geldanamycin(GA)对2株乳腺癌细胞株粘附基质胶能力的影响;侵袭小室重组人工基底膜侵袭实验检测HSP 90抑制剂GA对2株细胞的侵袭能力的影响;RT-PCR和Western blot实验检测2株细胞中HSP 90、HSP 70和HSP 27 mRNA及其蛋白质水平的表达差异。结果:GA能够明显抑制MDA-MB-435s和MDA-MB-231细胞粘附基质胶的能力(P<0.01);2株高转移性乳腺癌细胞株的侵袭能力比较无显著性差异(P>0.05),GA能够明显抑制2株乳腺癌细胞的侵袭能力,与对照组相比,MDA-MB-231细胞侵袭能力下降(P<0.05),MDA-MB-435s细胞的侵袭能力下降更为明显(P<0.01)。MDA-MB-435s细胞中HSP 90蛋白质和HSP 90αmRNA表达水平都明显高于MDA-MB-231细胞(P<0.01),MDA-MB-435s和MDA-MB-231细胞中HSP 90βmRNA表达水平没有显著差异(P>0.05);MDA-MB-231细胞中HSP 27的mRNA和蛋白质的表达水平均明显高于MDA-MB-435s细胞(P<0.01);MDA-MB-435s细胞中HSP 70蛋白质的表达水平与MDA-MB-231细胞没有显著性差异(P>0.05),HSP 70 mRNA表达水平低于MDA-MB-231细胞(P<0.05)。结论:HSP表达特性与乳腺癌细胞的粘附、侵袭能力相关;MDA-MB-435s细胞中HSP 90较高水平表达,MDA-MB-231细胞中HSP 27表达水平较高。Objective: To investigate expression and significance of 90, 70, 27 kDa heat shock protein (HSP90, HSP70, HSP27) in MDA-MB 435s and MDA-MB-231 breast cancer cell lines. Methods: The effects of HSP90 inhibitor,Geldanamycin (GA),on the adhesion to artificial basement membrane, Matrigel of breast cancer lines were assayed by MTT methods. The effect of GA on the invasive ability of breast cancer cells was determined by the Boyden chamber assay of invasion, which measures the ability of a ceil to invade through an artificial basement membrane. The mRNA and protein expression of HSP27, HSP70, and HSP90 was detected by RT PCR and Western blotting, respectively. Results: GA greatly inhibited the adhesion to Matrigel of MDA-MB-435s and MDA-MB-231(P〈0.01). The difference in invasion ability between MDA-MB-435s and MDA-MB-231 cells was not significant (P〉0.05). After treatment with GA, the invasion ability of MDA-MB-435s and MDA MB-231 cells were both significantly reduced(P〈0.01 ,P〈0.05), The inhibitory effect was greater in MDA-MB-435s cells compared with MDA- MB-231 eells(P〈0.01). The protein expression of HSP90 and the mRNA expression of HSP90αin MDA-MB-435s was higher than that in MDA-MB-231 cell (both P〈0. 01). The mRNA level of HSP90β had no signifcant difference between the two cancer cell lines (P〉0.05). The HSP27 protein and mRNA expression in MDA-MB-231 was higher than that in MDA-MB-435s cell (P〈0.01). The protein expression of HSP70 in MDA-MB-435s and MDA-MB-231 cells had no significant difference(P〉 0.05). The mRNA expression of HSP70 in MDA-MB-231 was higher than that of MDA-MB-435s cell(P〈0.05). Conclusion: HSP expression patterns were associated with the adhesive and invasive abilities of breast cancer lines. MDA-MB-435s cell had high expression level of HSP90 and MDA MB231 cell had high expression level of HSP27.
