摘 要:目的探讨给大鼠喂以低铁饲料辅以少量放血,快速建立缺铁性贫血的动物模型并观察动物营养状况及几种微量元素在脏器中的变化。方法将雌雄各半的SD大鼠随机分为两组(正常对照组:喂以正常饲料,饮用自来水;缺铁组:于塑料不锈钢笼内饲养,喂以低铁饲料,铁含量为8.01mg/kg,饮用去离子水)。每周测定血红蛋白浓度,当血红蛋白降低,确定模型建立后,每组随机取出8只大鼠,取其脏器,测定脏器系数(与体重比)及微量元素含量变化。结果缺铁性贫血大鼠的一般体态发生变化,体重明显下降。一个月后,缺铁性贫血大鼠组的血红蛋白浓度低至86.9g/L,其红细胞及全血铁指标也有显著的下降。活杀其中的8只大鼠,取出其心脏、肺脏、肝脏、脾脏、肾脏等器官测量脏器重量,计算出脏器系数值。缺铁组大鼠心脏和脾脏系数与正常组比较显著增高,差异明显,肝脏、肺脏、肾脏无明显差异。测定肝脏及大脑组织中微量元素铁、锌、硒、锰、镉、铅、铜等的含量。发现缺铁组大鼠肝脏及脑组织总铁含量明显低于正常组,锰的含量高于正常组,缺铁组肝脏铜高于正常组,脑锌含量低于正常组,其它几种元素变化不明显。结论给大鼠喂以低铁饲料辅以少量放血,可以成功建立缺铁性贫血的动物模型。其各项指标有明显的变化,尤其缺铁对脏器中微量元素含量有一定的影响。Objective To establish a rat model with iron deficiency anemia (IDA)by feeding the rat with a low iron diet and bleeding. To monitor the animal nutrition indexes and the change of related trace elements in internal organs. Methods SD rats were randomly divided into two groups (control and IDA group). The control group was fed with normal feed and water. The IDA group was fed with low iron (8 mg/kg) animal feed and distilled water. Low iron bait vessel was used in the IDA group. Hemoglobin was measured weekly till anemia was developed. When the IDA model was established, eight rats from each group were taken randomly and their organs were removed. The organ index (ratio between the weight of organ and the weight of the body) was measured. Concentrations of trace elements in the organs were also determined. Results In the IDA group, physical appearance of the rat, the behavioral change, and the body weight were dropped remarkably. The concentration of hemoglobin was also dropped to 86.9 g/L. The concentration of RBC and iron in whole blood dropped magnificently. The organ index of heart and spleen were higher in IDA group than in the control group. The concentration of Fe and Zn in brain and liver decreased obviously in IDA group, while the concentrations of Cu and Mn were higher in IDA group than in the control group. Conclusion A rat model with IDA was successfully established by feeding the rat with a low iron content diet and followed by bleeding. In this model, a number of indexes could be used to correlate with iron deficiency. Especially, the correlation between iron deficiency and the concentrations of some related trace elements in organs.
