Fmoc固相直接法合成Aβ_(1-15)肽疫苗及其免疫活性

摘　　要：【目的】探讨实验室合成Aβ1-15肽疫苗的方法,并对其进行鉴定。【方法】选取Aβ42主要包含B细胞抗原决定簇的片段Aβ1-15,采用8分支多重抗原肽系统,以Fmoc固相直接法合成MAPAβ1-15;用MAPAβ1-15疫苗免疫接种C57BL/6小鼠,ELISA检测血清特异性抗Aβ抗体。【结果】用Fmoc法直接合成方法成功制备MAPAβ1-15疫苗,质谱检测其主要离子峰的相对分子质量为15458,与理论相对分子质量15451十分接近,但还有其它不同分子质量的离子峰存在。以合成的MAPAβ1-15免疫C57BL/6小鼠后,可产生高滴度的特异性抗Aβ42抗体,第3次接种后抗体平均滴度为1:467±196,第5次免疫接种后,平均滴度为1∶4367±1120。对照组的血清抗体检测基本无明显变化。【结论】Fmoc固相直接法可成功合成出MAPAβ1-15,合成出的MAPAβ1-15具有很好免疫活性。[Objective] To explore how to synthesize the Aβ1-15 peptide vaccine and identify the quality and the immune activity of synthesized Aβ1-15 peptide vaccine. [Methods] To synthesize multiple antigenic peptide Aβ1-15 by Fmoc solid-phase pepfide directly synthesis. The molecular of synthesized MAP Aβ1-15 was analyzed by matrixassisted laser desorption/ionization time-of-flight mass spectrometric （MALDI-TOF MS）. C57BL/6 mice were immunized with synthesized Aβ1-15 vaccine. The specific anti-Aβ-antibody in the serum of immunized mice was identified by ELLSA. [Results] The molecular weight of main component in synthesized MAP Aβ1-15 was 15 458, and it was quite near standard molecular weight of 15 451. Besides the main component, there were still others of different molecular weight. High titer of anti Aβantibody was induced in C57BL/6 mice after inoculated with MAP Aβ1-15 , the average titer was 1：467±196 and 1：4 367±1120 after third and frith time inoculated, respectively. [Conclusion] MAP Aβ1-15 could be synthesized successfully by Fmoc solid-phase peptide directly synthesis. The synthesized complex has significant immune activity.
