金路捷对大鼠脑神经细胞损伤的保护作用

摘  要:目的建立体外培养大鼠脑神经细胞损伤模型,测定损伤前后不同时间LDH的水平,研究金路捷对脑皮质神经元的保护作用机制。方法原代培养新生Wistar大鼠的脑皮质神经细胞,建立体外脑神经损伤模型,通过测定神经细胞培养液中乳酸脱氢酶(lactatedehydrogenase,LDH)的含量变化观察模型对神经细胞的损伤作用。结果损伤组与对照组神经细胞损伤后LDH值比较差异均有显著性(P<0.05),证明损伤模型确实有效;本实验表明治疗组神经元LDH的水平与损伤组比较有差异(P<0.05),证明金路捷能够起神经元保护作用。结论损伤模型损伤前后各时间点LDH活力损伤组与对照组比较,表明本实验应用的损伤模型确实使细胞损伤。Objective To establish a model of brain nerve cell injury in vitro and examine the level of LDH at different times. This study also focused on understanding the mechanism of action for the protective effect of BDNF on cortical neurons. Methods To cultivate the rat cortical neurons and establish a model of brain nerve cell injury in vitro. According to the levels of LDH in cultivating medium at different times,detecting damage effect of the model. Results The concentration of LDH of the injured group had notable discrepancy compared to that of the control group (P 〈0.05) . It showed the feasibility of the model. The level of LDH of the injured group had discrepancy compared to that of the BDNF group (P 〈 0.05) . It showed both of the drugs could repair neurons. Conclusions The model is easy to perform and facilitate to observe the gradual changes of the injured neurons, which also can simulate the pathological mechanism of traumatic brain injury, and research the changes of the activity of the liquidity of the cell membrane.
