摘　　要：目的探讨人结直肠肿瘤组织中胃肠富集Kr櫣ppel样因子(KLF4)的表达及其意义。方法采用组织微阵列结合免疫组织化学染色和Westernblot检测经病理专家确认40例癌旁正常结直肠黏膜、7例腺瘤和77例结直肠癌组织中KLF4蛋白的表达,分析KLF4表达与临床病理因素间的关系。结果KLF4在结直肠肿瘤组织中的表达率为76.62%(59/77),其中弱阳性率53.25%(41/77),强阳性率为23.38%(18/77),癌旁形态学正常的结直肠黏膜可见KLF4高表达;强阳性率在正常黏膜、腺瘤、高分化、中分化癌中分别为57.5%、57.14%、37.5%、21.4%,低分化癌无强阳性表达,其表达率与肿瘤分化程度显著相关(Р<0.01)。此外,KLF4表达与患者性别、年龄、肿瘤位置、浸润深度、是否转移、大体类型以及临床Dukes分期无显著相关(P>0.05)。结论KLF4在结直肠肿瘤中表达下调,与其病理分级呈负相关,提示KLF4可能参与了结直肠肿瘤的发生及发展过程。Objective tal carcinoma and its investigate the expression of gut-enriched Krüppel like factor（ KLF4 ） protein in coloreccorrelation with clinicopathological feature. Methods Immunohistochemistry staining and Western blotting were performed to detect the KLF4 expression with the paraffin sections of tissue microarray, which contains 77 colorectal cancers, 40 normal mucosa and 7 adenoma. The relationship between the KLF4 expression and the clinicopathological parameters was analyzed . Results The positive rates of KLF4 protein expression in normal colorectal mucosa and adenoma both were 100%. Significant difference of KLF4 protein expression was found between normal mucosa and carcinomas （P 〈 0. 01 ）, which were further testified by Western blotting. KLF4 protein expression was inversely correlated with histological grade （ P 〈 0. 01 ）. No relationship was found between KLF4 protein and other clincopathological characteristics （P 〉 0. 05 ）. Conclusion The decrease of KLF4 expression in colorectal carcinoma is oppositely related to pathologic grade, which snqqests that KLF4 contribute to the development of colorectal carcinoma.
