摘  要:[目的]分析bcl-6基因5′端非编码区-1结构多态性及其蛋白在泌尿系统移行上皮癌及远端正常移行上皮组织中的表达情况.[方法]收集47例膀胱、肾脏及尿道移行上皮癌以及20例远端正常移行上皮组织蜡块,应用PCR-SSCP分析bcl-6基因5′端非编码区E 1.11,E 1.12区结构多态性,并应用免疫组织化学染色方法检测bcl-6蛋白表达情况.[结果]bcl-6基因5′端非编码区E 1.12的结构多态性多见于Ⅰ级移行上皮癌中.bcl-6蛋白在正常移行上皮组织中的表达阳性率较低,且规则地分布在表层;在移行上皮癌组织中bcl-6蛋白的表达率较高,且分布不规则.[结论]bcl-6基因5′端非编码区E 1.12区域的结构多态性及bcl-6蛋白的过度表达可能在移行上皮癌的分化和分级过程中起重要的作用,但两者无相关性.OBJECTIVE To analyze the 5' non-coding exon 1 of bel-6 gene and its protein expression in transitional cell carcinomas (TCCs) and normal transitional epithelial cells adjacent to the TCCs. METHODS The paraffin blocks of 47 transitional cell carcinomas of the urinary bladder, kidney, urethra, and 20 normal transitional epithelial tissues adjacent to the TCCs were investigated by polymerase chain reaction-single stranded DNA conformation polymorphism (PCR-SSCP) and immunohistoehemieal methods. RESULTS The positive ra-tio of bcl-6 conformation polymorphism at exon 1.12 in grade I was significantly higher than that in grade Ⅰ and grade Ⅲ of TCCs. Bcl-6 protein had a low positive rate in normal transitional epithelium, and the positive cases showed regularly expression pattern in the upper layer of normal transitional epithelia, whereas, bcl-6 protein was irregularly and heavily stained in the nucleus of the TCCs. CONCLUSION Bcl-6 gene conformation polymorphism at exon 1.12 and the over-expression of bel-6 protein may play some roles in the differentiation and histopathologie grade of TCC, but the over-expression of bel-6 protein was not related with the bcl-6 gene conformation polymorphism at 5' non-coding exon 1.
