摘  要:目的:观察H2O2、SOD和膜固思达对人胃癌细胞系MGC-803细胞P53、P21ras蛋白表达的影响,探讨氧自由基在胃癌发生发展中的作用。方法:通过对MGC-803细胞培养并给予一定浓度的H2O2、SOD和膜固思达以调节细胞内氧自由基水平,采用硫代巴比妥酸反应物质法检测细胞内氧自由基中间产物丙二醛(MDA)的含量,应用免疫组织化学技术检测细胞的P53、P21ras蛋白表达的情况。结果:H2O2使细胞内氧自由基水平升高,P53、P21ras蛋白表达增强(P<0.01),SOD和膜固思达降低细胞内氧自由基水平,P53、P21ras蛋白表达减弱(P<0.01)。结论:H2O2、SOD和膜固思达可通过改变MGC-803细胞内氧自由基水平而影响其P53、P21ras蛋白的表达。Objective: To observe the effects of H2O2 ,SOD and Mucosta on P53 and P21ras protein expressions in human gastric cancer cell line MGC-803 and explore the role of oxygen free radical (OFR)in gastric cancer development. Methods: MGC-803 cells were cultured and given H2O2,SOD and Mucosta in some concentration to regulate the level of OFR in the MGC-803 cells.The content of malondialdehyde (MDA) in the MGC-803 cells was measured as middle product of OFR by TBARS method. Immunohistochemical staining (IHC) was used to detect the expressions of P53 and P21ras protein in the MGC-803 cells. Results: Giving H2O2 to make the level of OFR in the MGC-803 cells rise, the expressions of P53 and P21ras protein were promoted (P〈0.01) .The level of OFR was decreased by giving SOD and Mucosta and so were the expressions of P53 and P21ras protein (P〈0.01). Conclusion: H2O2 ,SOD and Mucosta can influence on the expressions of P53 and P21ras protein by changing the level of OFR in the MGC-803 cells.
