摘 要:在使用扩孔剂间三甲苯的条件下,对新型大孔纳米材料MCM-41进行了制备,并对该材料作为载体固定化木瓜蛋白酶进行了研究.X射线衍射和氮气吸附表征结果表明:材料内部孔的排列高度有序且孔径集中分布在6.3nm附近.用该材料作为载体固定化木瓜蛋白酶的结果表明:在给酶量、戊二醛质量分数、时间、温度和pH值分别为20mg/g,0.75%,2h,10~20℃和7.0的条件下,固定化酶的回收率为55%左右,效果最佳.The present work dealt with the preparation of nanoporous material MCM-41 with mesitylene being a swelling agent as well as the immobilization of papain on the as-synthesized material with the aid of a coupling agent, glutaraldehyde. It was inferred from the diffraction data and the nitrogen sorption isotherms of the as-synthesized material that the material exhibited a highly ordered arrangement of uniform nanopores whose dimensions were determined to be around 6.3 nm. The optimum conditions for the immobilization were as follows: the concentration of native free enzyme, glutaraldehyde concentration, time, temperature and pH were 20 mg/g, 0.75%, 2h, 10-20℃ and pH 7.0, respectively. Under the optimum conditions, the activity yield ( % ) of the immobilized enzyme was around 55 %.
