摘  要:目的观察抗CD81抗体EAT2和H121对培养大鼠视网膜神经胶质细胞(RGC)增生活性的影响。方法分别将2μg/ml、10μg/ml抗CD81抗体EAT2和H121加入体外培养SD大鼠RGC中,7d后以MTT法测定细胞生长抑制率。结果加入抗CD81抗体后RGC增生活性下降,H121对RGC的细胞生长抑制率可达50.37%。结论抗CD81抗体EAT2和H121可以抑制培养大鼠RGC的细胞增殖活性。Objective As a member of tetraspanins superfamily, CD81 is proved to be involved in the inhibition of cellular mitotic activity, including mieroglial cell and astroeytes from brains of rats. The present study is to find out the effect of CD81 on the growth of rat retinal glial cells. Methods Retinal glial cultures were established from retina of 3-day-old Sprague-Dawley rats. The third passage of cells were used for assay. Various concentrations(2 μg/ml and 10 μg/ml)of two types of anti-CD81 antibody(EAT2 and H121) were added to culture medium respectively. Control cultures were treated with non-immune IgG at corresponding concentration. Their effect on the growth of these cells was determined by MTT eolorimetric assay after 7 days, and the inhibition rates were calculated accordingly. ResuIts As compared to the control, the OD values of EAT2 treated cultures were significantly reduced at the concentration of 2 μg/ml (P = 0.015 288) and 10 μg/ml (P = 0.001 087), while that of the H121 group was depressed even more prominently at both 2 μg/ml (P = 0.000 509) and 10 μg/ml (P = 0.000 251). The inhibition rate of retinal glials was 20.04% and 25.90% in EAT2, 40.60% and 50.37% in H121 group at concentration of 2 μg/mland 10 μg/ml respectively. Conclusion The combination of CD81 with its antibody shows close relationship to the growth activity of rat retinal glial cells in vitro.
