实验性青光眼大鼠血清中HSP27抗体水平检测及意义

摘　　要：目的观察实验性青光眼大鼠血清中热休克蛋白27(HSP27)抗体水平改变,分析眼压与抗体水平的关系。方法将55只Wistar大鼠随机分为高眼压组(25只)s、ham(假手术)组(25只)及正常组(5只)。采用电凝鼠眼巩膜表面至少三组静脉及角膜缘周围血管,减少房水静脉回流建立鼠眼青光眼模型。分别于术后1、2、3、4及8周处死大鼠,处死前测眼压,抽血2ml供酶联免疫吸附测定使用,分析针对视网膜中HSP27蛋白抗原产生的血清中相应抗体水平。结果进行统计学分析。结果高眼压组右眼术后眼压升高,术后1周眼压为(30.43±5.88)mmHg,此后眼压基本稳定。术后高眼压组右眼眼压与术前、左眼及sham组右眼比较,差异有统计学意义(P<0.01)。高眼压组血清中抗HSP27抗体水平术后1周轻度升高(P>0.05),从术后第2周直至8周,血清中HSP27抗体含量迅速升高,并稳定于较高水平,术后2、3、4及8周分别与各sham组、正常组及术后1周比较,差异有统计学意义(P<0.05)。结论伴随眼压升高出现的血清中HSP27抗体水平升高可能在青光眼视神经损害中具有重要作用。Objective To detect the levels of antibody against heat shock protein 27（HSP27） in serum in experimental rat glaucoma model and study the relationship between intraocular pressure （IOP）and the levels of anti-HSP27 antlbody.Methods 55 Wistar rats were randomly divided into the ocular hypertension,Sham and normal groups.IOP was elevated by electroccagulation of at least 3 episcleml and limbal veins in the right eye of each rat in the ocular hypertension gronp.At 1 week（w） ,2w,3w,4w and 8w after opera- tion,lOP was mcasured.The Serum anti-HSP27 antibody was detected by means of enzyme-linked immunesorbent assay.Results Electrocaagulation treatment caused a significant IOP increase of the right eyes in ocular hypertension group over the baseline, contra-lateral eyes and sham eyes（ P 〈 0.01 ） .At lw, IOP was 30.43 ± 5.88 mmHg（ 1KPa = 7.5 mmHg）, and stabillzed thereafter to 8w.Compared with sham and normal groups,serum anti-HSP27 antibody was slight high at lw（ P 〉 0.05）and significant increase at 2w,3w,4w and 8w （ P 〈 0.05）in ocular hypertension group. Conclusion The enhanced level of anti-HSP27 antibody in serum with lOP increase in experimental rat glaucoma model might play an important role in glaucomatoas optic neuropathy.
