摘　　要：目的探讨环氧化酶-2抑制剂塞来昔布(celecoxib)对人卵巢癌细胞系A2780细胞增殖与凋亡的影响。方法采用MTT法观察塞来昔布对A2780细胞的增殖抑制情况;流式细胞仪测定塞来昔布对A2780细胞周期的影响;并用荧光显微镜观察凋亡细胞的形态。结果塞来昔布对A2780细胞的增殖具有抑制作用,并显示表现出明显的时间和剂量的依赖性;塞来昔布使A2780细胞发生细胞周期的G1期阻滞,细胞凋亡增加,并且在荧光显微镜下可观察到凋亡细胞。结论塞来昔布抑制A2780细胞增殖并促进细胞凋亡,有可能成为卵巢癌化疗的有效药物。Objective To investigate the effects of celecoxib on proliferation and apoptosis of the human ovarian carcinoma cell line A2780. Methods Proliferation was assessed by flow cytometry and MTT assay ; apoptosis was detected by flow cytometry and fluorescent microscope. Results Proliferation of A2780 cell line was significantly inhibited by celecoxib in some time and dosage dependent manner; celecoxib caused G1 arrest of A2780 cell line associated with increase of the DNA content in G1 stage and decrease of the DNA content in S and G2/M stage, and caused apoptosis. Conclusions Celecoxib inhibited A2780 cell proliferation, caused its proliferation to arrest in G1 stage and apoptosis.
