摘  要:目的研究视神经钳夹伤后凋亡相关基因Bcl-2和Bax在大鼠视网膜上的表达,探讨Bcl-2和Bax在视神经钳夹伤后视网膜神经元凋亡中的作用。方法将雄性LongEvans大鼠50只,随机分成实验组和对照组,每组25只大鼠。建立大鼠视神经钳夹伤模型,实验组和对照组按视神经夹伤后不同时间分为伤后1 d、3 d、5 d7、d1、4 d 5个时间点,每个时间点5只动物。处死动物后取材,制备大鼠视网膜石蜡切片,进行免疫组化SABC染色,以检测视网膜Bcl-2和Bax的表达。结果视神经钳夹伤后视网膜Bcl-2的表达下降,而Bax表达仍为不变的水平。结论视神经钳夹伤后视网膜Bcl-2表达的下降,导致了视网膜神经元的凋亡。Objective To investigate the expression of apoptasis-related genes (Bcl-2, Bax) in the rat retina after crushaing the optic nerve (ON) with microforceps, and to investigate the role of these genes in retinal cell apoptosis after ON crushing.Methods Male Long Evans rats were randomly divided into test groups and control groups (n = 25, per group). The experimental model of ON crushing with microforceps was established. Test groups and control groups were both divided into 5 groups according to the amount of time that had elapsed after ON crushaing (1 d, 3 d, 5 d, 7 d, and 14 d). The rat were sacrificed on postoperative days 1, 3, 5, 7 and 14. Eyes were dissected and immediately frxed in eye cups without the cornea or lens for 48 hours in 4% paraformaldehyde at 4℃, then paraffin sections of the retina were cut, Retinal paraffin sections were stained by immunohistochemical method to detect the expression of apoptosis-related genes Bcl-2 and Bax in the retinas. Results There was a strong expression of Bax in the retina after ON crushing while the expression of Bcl-2 had gradually weakened. Conclusion The decrease in Bcl-2 expression may lead to the apoptosis of retinal neurons after ON crushing with microforceps.
