摘  要:目的:寻找喉癌新的相关基因,阐明喉癌发生的分子机制,为喉癌的基因诊治提供研究的靶基因。方法:应用mRNA差异显示方法对喉癌患者的癌旁、癌及颈部转移淋巴结组织差异cDNA进行了筛选、克隆和测序,并对其进行了鉴定和同源性分析。结果:筛选出7个差异cDNA片段,克隆并鉴定了其中两个与喉癌发生相关的cDNA片段,它们与多种肿瘤相关基因的cDNA序列同源。结论:克隆的两个cDNA片段可能是喉癌新的相关基因cDNA片段,全长cDNA克隆及深入研究其功能有助于发现喉癌新的相关基因。OBJECTIVE:To investigate novel genes related to laryngeal carcinogenesis. METHODS: cDNA from cancerous tissue, adjacent normal laryngeal tissues (PANL) and metastasis tissue in the same patient with laryngeal cancer were compared by mRNA differential display (mRNA-DD). Differentially expressed cDNA fragments were cloned to pGEM-T vector, sequenced and identified by semi-quantitiy RT-PCIL The cDNA sequences were compared to those from internet by Blast. RESULTS: Seven differentially expressed cDNA fragments were found, two of which were cloned and sequenced. RT-PCR result showed that A fragment was highly expressed in normal laryngeal tissue and D fragment was expressed mainly in laryngeal carcinoma, which implied that A and D fragments might be parts of a novel candidate tumor suppressor gene and candidate oncogene, respectively. The homologous analysis demonstrated that the nucleotide sequences of the two cDNA fragments were similar to those cloned from many tumors. The amino-acid sequence of A were partly homologous to those of mei2-1ike protein and decay accelerating factor. CONCLUSIONS: The two cloned cDNA fragments might participate in the laryngeal carcinogenesis. Further research will help to discover novel genes related to laryngeal carcinogenesis and understand its molecular mechanism better.
