甲状腺素受体β1在胃癌组织中的表达及其意义

摘　　要：目的:研究甲状腺素受体β1(thyroidhormonereceptorβ1,THRβ1)在胃癌组织中的表达及其与临床病理因素之间的相互关系。方法:选用61例胃癌组织和10例癌旁正常组织,应用RTPCR琼脂糖凝胶电泳溴化乙锭染色法检测标本中THRβ1mRNA表达的变化,并结合患者临床病理指标进行分析。结果:THRβ1在61例胃癌组织中,20例表达呈阳性,阳性表达率为32.6%(20/61);而在10例癌旁正常胃组织中9例表达呈阳性,阳性表达率为90%(9/10)。在胃癌组织中的表达率明显低于邻近正常胃黏膜,P=0.002。THRβ1mRNA的低表达与患者的年龄、性别、肿瘤所在部位、肿瘤的BORRMANN分型、组织学类型、分化程度无关,P>0.05。与肿瘤大小,腹膜转移、肝转移、有无浆膜浸润、淋巴结转移、TNM分期、远处转移有关,P<0.05。结论:THRβ1mRNA的低表达在胃癌的发生中起一定作用,并且与胃癌的侵袭转移有关。OBJECTIVE： To explore the relationship between THRβ1 mRNA expression and clinicopathological parameters in human gastricancer. METHODS： RT-PCR was used to detect the expression of THRβ1 mRNA in 61 human gastric carcinomas and 10 adjacent normal gastric tissues. RESULTS： A positive rate of THRβ1 mRNA was 32.6% in 61 human gastric carcinomas and 90% in 10 adjacent normal gastric tissues. The expression level was lower in gastric carcinomas than in adjacent normal gastric tissues, P = 0. 002. The expression was not correlated with patients＇ age, sex, tumor location, histologic types, differention. The expression level was lower in late-stage tumors （stage Ⅲ and Ⅳ） than in early-stage tumors （stage Ⅰ and Ⅱ ）, P=0. 001 ; in tumors with invasion to serssa than those without serosal invasion, P=0. 001 ; in tumors with lymph node metastasis than those without lymph node metastasis, P= 0. 007; and in large-sized tumors than in small-sized ones,P=0. 009. CONCLUSION： THRβ1 might play an important role in the development of invasion and metastasis of gastric cancer.
