摘  要:目的:研究细胞凋亡相关基因Fas和FasL在胰腺癌组织中的表达及意义。方法:采用免疫组织化学技术观察Fas及FasL蛋白的表达,用原位末端标记法检测细胞凋亡。结果:40例胰腺癌中Fas和FasL阳性30例,阳性率76·5%;用于对照的10例癌旁正常胰腺组织中Fas和FasL阳性1例,阳性率10%,Fas和FasL在胰腺癌组织中表达高于对照组,χ2=14·3463,P=0·0002;在正常胰腺组织连续切片中未发现TdT酶标记的DNA3/-OH末端,提示无细胞凋亡;在胰腺癌标本中用TdT酶标记的DNA3/-OH末端分析连续切片表明,大多数癌细胞未发生细胞凋亡,在40例胰腺癌中仅有3例发现约10%的癌细胞凋亡。结论:Fas和FasL参与了大部分胰腺癌的凋亡调变,胰腺癌在发生分化过程中,可能部分获得了逃逸Fas介导的凋亡功能。OBJECTIVE: To study the expressions and significance of proapoptosis gene Fas and FasL in pancreatic cancer. METHODS: Immunohistochemical technique was used to observe Fas and FasL expressions in normal and cancerous tissues. DNA 3/-OH end labeling was used to detect apoptotic cells. RESULTS: Fas and FasL immunoreactivity was detected in 30 cases from 40 patients with pancreatic cancer and 1 cases from 10 normal pancreatic tissue. Expressions of Fas and FasL in PC were higher than those in mormal pancreatic tissue (x^2 = 14. 346 3, P=0. 000 2); apoptotic signal was present in approximately 10% of the tumor cells in only 3 of 40 cancer samples. CONCLUSION: Fas and FasL were expressed in most of PC. Fas and FasL may be involved in the modulation of apoptosis in PC, but PC may have partly adapted the function of evasion Fas-mediated apoptosis.
