摘 要:目的探讨色素上皮衍生因子(PEDF)对氪激光诱导的BN大鼠脉络膜新生血管(CNV)的抑制作用。方法16只BN大鼠随机分成2组进行单眼氪激光视网膜光凝。光凝后第1周,一组玻璃体内注射重组人色素上皮衍生因子(rhPEDF);另一组玻璃体内注射平衡盐溶液(BSS)作为对照组。光凝后第2周和第4周,分别对两组动物进行眼底血管荧光造影检查(FFA)及光凝区Ⅷ因子相关抗原(FⅧR∶Ag)免疫组织化学检测,分析比较脉络膜新生血管形成密度。结果光凝后第2周和第4周,FFA检查同一时间点rhPEDF组荧光素渗漏平均密度低于对照组。FⅧR∶Ag免疫组织化学检测结果经半定量分析,对照组由第2周至第4周可见光凝区内FⅧR∶Ag阳性染色密度增加,而rhPEDF组光凝区内的FⅧR∶Ag阳性染色密度无增加;视网膜光凝后第2周和第4周,rhPEDF组光凝区FⅧR∶Ag阳性染色密度分别低于对照组。结论玻璃体内注射PEDF对氪激光诱导的BN大鼠CNV具有抑制作用,PEDF有可能成为一种有临床应用前景的CNV抑制剂。Objective To investigate the inhibitory effects of pigment epithelium-derived factor(PEDF) on the formation of choroidal neovascularization (CNV) induced by krypton laser in BN rat. Methods Two groups of 16 BN rats were photocoagulated by krypton laser in single eye. One week after photocoagulation, one group of 8 rats received intravitreal injections of 2 μg/10μl rhPEDF, and the other group of 8 rats received intravitreal injections of 10 μl balanced salt solution(BSS) as control group. Two weeks and 4 weeks after photocoagulation, fundus fluorescein angiography(FFA) was performed, and the expression of FWR: Ag was observed by immunohistochemical method. Results FFA showed less fluorescein leakage in the PEDF group than that in the control group 2 weeks and 4 weeks after photocoagulation respectively. From 2 weeks to 4 weeks after photocoagulation, the expression of FⅧR: Ag showed increase in the control group, but no increase in the PEDF group; the expression of FⅧR:Ag in PEDF group was less than that in the control group at the same phase. Conclusions Intravitreal PEDF injection can inhibit CNV induced by krypton laser in BN rat, and PEDF may be an effective inhibitor for the treatment of CNV.
