摘 要:目的探讨肿瘤病人拟放射样化合物暴露后DNA的修复能力。方法采集20名不同类型肿瘤病人和20名非肿瘤病人的淋巴细胞,用浓度为20μg/ml的博来霉素(Bleomycin)作用30 min后,再经15 min修复,采用彗星试验检测Bleomycin暴露前后DNA损伤程度来评价DNA修复能力。结果Bleomiycin攻击试验中尾长和尾相所测肿瘤组平均DNA修复能力分别为79.42%、75.97%,明显低于对照组(92.41%,90.69%),P<0.01。结论Bleomycin攻击试验结合彗星试验可以快速检测DNA修复能力,发现肿瘤病人DNA修复能力明显低于非肿瘤病人。[ Objective] To detect the DNA repair capacity of cancer patients after the exposure to radiomimetie compounds. [ Methods ] The peripheral lymphocytes of 20 cancer patients of different kinds and 20 healthy people were collected and treated with Bleomycin (20μg/ml) for 30 minutes and repaired for 15 minutes. Comet assay was applied to detect the DNA damage before and after BLM exposure to assess the individual DNA repair capacity. [Results] The average DNA repair capacity of cancer patients by tail length and tail moment in Bleomycin challenge test were 79.42% and 75.97% respectively, which were significantly lower than that of control group (92.41% and 90.69 % ) ( P 〈 0.01 ). [ Conclusion] Bleomycin exposure test combined with comet assay can rapidly assess the DNA repair capacity. It was found that the DNA repair capacity of cancer patients was significantly lower than that of the control group.
