摘 要:目的研究大鼠皮肤损伤愈合过程中血管内皮细胞生长因子(VEGF)和促血管生成素-1(Ang-1)表达的动态变化,阐明其在组织修复血管再生中的作用。方法在大鼠背侧切开皮肤全层建立大鼠创伤模型,不同时问后活体取材,应用HE染色观察组织病理形态学变化;应用免疫组化SP法检测VEGF和Ang-1的蛋白表达。结果伤口组织病理学检查可见,毛细血管在伤后第3d即见增多,伤后第7d达高峰,随后逐渐减少。血管内皮细胞胞浆VEGF蛋白表达在伤后第1d呈阳性。伤后第3d呈强阳性,伤后第5d呈阳性,而于伤后第7d阳性程度逐渐减弱。伤后第1~14d VEGF蛋白表达与对照组比较,有非常显著性差异(P〈0.01)。血管内皮细胞胞浆Ang-1蛋白表达在伤后第1d部分呈阳性表达,部分呈弱阳性表达,在伤后第3d呈阳性表达。伤后第5、7d呈强阳性、阳性表达。随后阳性程度逐渐减弱。伤后第3~21d Ang-1蛋白表达与对照组比较,有非常显著的统计学差异(P〈0.01)。结论VEGF和Ang—1在血管内皮细胞中的表达强度与组织损伤愈合程度密切相关,在创伤愈合过程中互相协调、促进并调节血管肉芽组织的生长和成熟。Objective To observe the expression of vascular endothelial cell growth factor(VEGF) and angiopoietin-1 (Ang-1) and illuminate their effects of the capillary regeneration during skin wound healing in rats. Methods A full-thickness incision was made on the dorsal skin. The specimens were obtained at different times after wound. The pathomorphology changes of the wound tissue were observed by HE staining. The expression of VEGF and Ang-1 in the wound tissue was detected by SP immunohistochemical techniques. Results The number of capillary began to increase at the 3rd day, and reached the peak at the 7th day and decreased gradually after the 7th day of the wound. The VEGF protein was positive in plasma of endothelial ceils at the 1st day, and strong positive at the 3rd day, and positive at the 5th day in these cells and decresed at the 7th day after wound. Compared with normal tissue, they showed significant difference from the 1st day to the 14th day after wound(P 〈 0.01 ). The Ang-1 protein was positive in the plasma of some endothelial cells or slightly positive part- ly at the 1st day, and positive at the 3rd day, and strong positive partly or positive partly at the 5th and the 7th day in these cells, and then decreased after the 7th day after wound. Compared with normal tissue, they showed significant difference from the 3rd day to the 21st day after wound (P〈 0.01).Conclusions There is a time-related expression of VEGF and Ang-1 in endothelial cells during wound healing of the rat skin. They promote and regulate vascularization of granulation tissue in coorperation manner during wound healing.
