摘 要:目的应用骨形态计量学方法观察雄激素对去睾丸大鼠不同结构骨组织骨代谢的影响。方法30只4月龄SD雄性大鼠,随机分成年龄对照组(A组)、去睾丸组(B组)和去睾丸加睾丸酮组(C组)。A组和B组每日给予生理盐水5m·lkg-1,C组每日给予甲基睾丸酮片1.8mg·kg-1,灌胃给药,持续90d。实验结束后,处死全部大鼠,取胫骨上段和中段进行不脱钙骨包埋切片,应用计算机自动图象分析系统进行骨组织形态计量学分析。结果与A组比较,B组大鼠胫骨上段骨量下降(%Tb.Ar-67.7%,P<0.05),骨形成和骨吸收参数都增加;胫骨中段皮质骨静态参数截面总面积、髓腔面积等无明显变化,动态参数骨外膜骨形成降低,骨内膜的骨形成和骨吸收均有增加趋势。与B组比较,C组大鼠胫骨上段的骨量增加(%Tb.Ar+79.8%,P<0.05),骨高转换率降低;胫骨中段皮质骨骨外膜的骨形成增加,骨内膜的骨吸收降低(P<0.05)。结论雄激素替代疗法可以防止大鼠由于雄激素水平下降造成的松质骨骨量丢失;并能正常的皮质骨结构,但作用相对缓慢。Objective To study the effects of testosterone on different part of skeletal metabolism in OBX rats by bone histomorphometry. Methods Thirty four-month-old Sprague Dawley male rats were randomly divided into three groups: age control group(group A), OBX group(group B) and OBX with methyltestesterone group(group C). Group A and B were administered with saline 5ml·kg^-1·d^-1,group C were given with methyltestosterone 1.8mg·kg^-1·d^-1.After 90 days, all rats were killed and their proximal tibia and tibial diaphysis were processed undeealcified sections and measured by automatic image-analysis system for bone histomorphometry. Results Compared with A group, OBX rats of group B had less bone mass(%Tb. Ar-67.7%, P〈0.05)and higher bone formation and resorption rate in proximal tibial metaphysic (PTM). static parameters of cortical bone such as total tissue area and marrow cavity changed little but dynamic parameters as periosteal bone formation decreased(P〈0.05) and endosteal bone turnover inclined to increased, bone mass of PTM more improved (%Tb.Ar+ 79.8%,P〈0.05) and higher bone turnover reduced in group C than in group B. Testosterone also could elevate periesteal formation and endesteal bone resorption. Conclusions Testosterone substitute therapy can prevent the trabecular bone loss, and maintained the cortical bone's normal tructure of rats with low testosterone.
