摘 要:目的研究强力霉素对人胃癌细胞SGC-7901增殖的影响及其机制。方法用MTT法和免疫组织化学法分别检测SGC-7901细胞的增殖及基质金属蛋白酶-2(matrix metalloproteinase-2,MMP-2)蛋白表达水平。结果强力霉素作用于SGC-7901的抑制效应呈浓度和时间依赖性,SGC-7901经强力霉素处理后MMP-2的表达,不同浓度间有显著性差异(P<0.01)。结论强力霉素对胃癌细胞SGC-7901有抑制增殖的作用,其机制可能与下调MMP-2蛋白表达有关。Objective To study the effect ofdoxycycline on human gastric cancer cells SGC-7901 proliferation in vitro and its mechanism. Methods The proliferation of SGC-7901 was detected by MTT assay, the matrix metalloproteinase-2(MMP-2) protein level was measured with immunohistochemistry. Results Doxycycline could inhibit the proliferation of human gastric cancer cells SGC-7901. The rate of inhibition increased with time and dosage dependence. Immunohistochemical results demonstrated that doxycycline could decrease the MMP-2 protein expression(P 〈 0.01)in a concentration-dependent manner. Conclusion Doxycycline could inhibit the proliferation of SGC-7901, possibly by decreasing MMP-2 protein expression.
