摘 要:目的研究肿瘤原发灶和转移灶的基因表达差异,并采用生物信息学方法对1条卵巢癌肝转移灶低表达基因C14orf106进行初步分析。方法分别制备卵巢癌原发灶和肝转移灶组织标本mRNA,线性扩增后与人类寡核苷酸芯片杂交,通过信号扫描、处理后获得两者的表达差异基因。并用生物信息学方法对1条无功能研究的新基因C14orf106进行初步分析,预测了它的基因结构、染色体定位、编码蛋白质的理化性质、亚细胞定位、蛋白质功能域等信息。并对多物种中的相似性蛋白进行了系统进化分析。结果人类寡核苷酸芯片发现了共272条差异表达基因。对转移负相关新基因C14orf106的上述性质进行了有效的预测,基本明确了该基因编码蛋白为一核蛋白,可能参与了肿瘤转移抑制基因的转录活化及促进表达。结论生物芯片技术是一种研究肿瘤转移基因表达差异的有效的高通量研究方法。通过生物信息学分析,表明新基因C14orf106是一个有肿瘤转移研究价值的新靶点。[Objective] To explore the differential expressed genes in tumor between in situ and metastasis, and perform an initial bioinformaties analysis on a novel gene, C14orf106, which is low expressed in hepatic metastasis. [Method] The mRNA of the tissues from the in situ site and hepatic metastasis were extracted respectively and hybridized with human oligo mieroarray after amplification, and the differentially expressed genes were obtained. Initial bioinformaties analysis was performed on a novel gene named C14orf106 which had no functional researches at present. Its gene structure, genomie localization, the physical and chemical characteristics of the putative protein, subeellular localization, functional domain and ere were predicted, and the systematic evolution analysis was performed on the similar proteins among several species. [Result] Using oligo mieroarray, many differentially expressed genes were uncovered. Efficient bioinformaties analysis had fundamentally identified that C14orf106 protein was a nuclear protein located on endoplasmie retieulum, and might involved in the transcriptional activation and expression promotion of anti-metastasis genes. [Conclusion] Mieroarray is an efficient and high throughout strategy for the detection of differential expressed genes. And C14orf106 is thoughtto be a potential target for tumor metastasis researches by bioinformaties analysis.
